目的:对地蚕(Stachys geobombycis)乙酸乙酯部位和正丁醇部位进行系统的化学成分进行研究。方法:将干燥的地蚕块茎粗粉10kg用95%乙醇回流提取3次,合并提取液,减压浓缩得到浸膏,将浸膏加适量水分散,依次用石油醚、乙酸乙酯及正丁醇分别进行萃取,得到石油醚部位、乙酸乙酯部位和正丁醇部位。对其乙酸乙酯部分和正丁醇部分通过硅胶色谱法,LH-20羟丙基葡聚糖凝胶(Sephadex LH-20)柱色谱和半制备HPLC等技术手段进行分离纯化,分离得到单体化合物。依据各化合物的理化性质和谱学数据鉴定其化学结构。结果:从地蚕的乙酸乙酯和正丁醇提取物中分离得到12个化合物,分别为麦角甾苷(1),异类叶升麻苷(2),对羟基苯甲酸(3),latifolicinin C(4),papuline(5),亚油酸甲酯(6),α-亚麻酸(7),3,4-二羟基苯甲醛(8),尿嘧啶核苷(9),methyl 3,4-dihydroxy-benzoate(10),β-谷甾醇(11),豆甾醇(12)。结论:化合物5,10,11为首次从该属中分离得到,化合物1~4,6~9,12为首次从地蚕中分离得到。
Objective: To study the systematic chemical components of ethyl acetate and n-butanol in Stachys geobombycis. Methods: 10kg of dried silkworm tuberous meal was extracted with 95% ethanol by refluxing for three times. The extracts were combined and concentrated under reduced pressure to obtain an extract. The extract was dispersed with appropriate amount of water and washed with petroleum ether, ethyl acetate and n-butyl acetate Alcohol were extracted, respectively, to obtain petroleum ether, ethyl acetate and n-butanol parts of the site. The ethyl acetate fraction and the n-butanol fraction were separated and purified by silica gel chromatography, Sephadex LH-20 column chromatography and semi-preparative HPLC, and the monomer compounds were isolated . According to the physical and chemical properties of each compound and spectroscopic data to identify the chemical structure. Results: Twelve compounds were isolated from the ethyl acetate and n-butanol extract of silkworm silkworm, which were acteoside (1), isoacteoside (2), paraben (3), latifolicinin C 4, papuline 5, methyl linoleate 6, α-linolenic acid 7, 3,4-dihydroxybenzaldehyde 8, uridine 4, methyl 3,4- dihydroxy-benzoate (10), β-sitosterol (11), stigmasterol (12). Conclusion: Compounds 5, 10 and 11 were isolated from this genus for the first time. Compounds 1 ~ 4,6 ~ 9,12 were isolated from silkworm for the first time.
