目的:考察合成冰片对复方中药湿疹乳膏体外释放和透皮吸收行为的影响,探讨该复方组成改良的可行性。方法:以离体BALB/c裸小鼠皮肤为渗透屏障,采用改良Franz垂直扩散池,比较含有合成冰片的复方中药湿疹乳膏和不含合成冰片的复方中药湿疹乳膏的透皮吸收效果,初步评价合成冰片的作用。采用HPLC测定透皮接收液中苦参碱的含量,流动相乙腈-水(含0.05%三乙胺,磷酸调p H 3.5)(2∶98),检测波长220 nm。结果:含合成冰片乳膏中的苦参碱累积渗透量在给药后的各时间点均高于无冰片乳膏,在12 h体外经皮渗透达到稳态。含合成冰片12 h累积渗透率6.37%,不含冰片的则为4.99%,二者具有显著性差异。结论:合成冰片对复方中药湿疹乳膏中苦参碱的透皮吸收具有一定促进作用,可在原复方处方中改良加入合成冰片。
Objective: To investigate the effects of synthetic borneol on the in vitro release and transdermal absorption of Compound Chinese Herbal Eczema cream, and to explore the feasibility of improving the composition of the compound. Methods: The skin permeability of BALB / c nude mice was used as the permeation barrier. The improved Franz vertical diffusion cell was used to compare the transdermal absorption of the compound traditional Chinese medicine eczema cream containing synthetic borneol and the compound traditional Chinese medicine eczema cream without synthetic borneol. Preliminary evaluation of the role of synthetic borneol. The content of matrine in transdermal receiving solution was determined by HPLC. The mobile phase consisted of acetonitrile-water (containing 0.05% triethylamine and phosphoric acid) (2:98). The detection wavelength was 220 nm. Results: The cumulative infiltration of matrine in the synthetic borneol cream was higher than that of the non-borneol cream at various time points after administration, and reached the steady state after 12 h of transdermal penetration. The accumulative permeability of synthetic borneol at 12 h was 6.37%, while that of borneol at 4.99%. There was a significant difference between the two. CONCLUSION: The synthetic borneol can promote the transdermal absorption of matrine in the compound traditional Chinese medicine eczema cream, and can be modified into the original borneol in the prescription of original compound.
