目的 研究放射性同位素局部照射对自体移植静脉段内膜增生反应的影响及其可能机理。方法 建立 4 0只大鼠自体静脉移植模型 ,随机分成对照组 (单纯自体静脉移植组 )、8、18、36Gy组 (按不同32 P内膜照射剂量 ) ,每组 10只。 2周后取材固定 ,行HE染色、Verhoeff弹力纤维染色、增殖细胞核抗原 (PCNA)免疫组化测定 (SP法 )、计算机图像分析仪测量移植血管内膜厚度。结果 移植静脉段的内膜平均厚度 :对照组与 8Gy组差异无显著性 (P >0 0 5 ) ,但高于 18、36Gy组(P <0 0 5 ) ,18Gy组与 36Gy组间差异无显著性 (P >0 0 5 ) ;PCNA阳性片示 ,18、36Gy组VSMC增殖较对照组受到明显抑制 (P <0 0 1) ,两组差异无显著性 (P >0 0 5 )。 8Gy组较对照组差异无显著性(P >0 0 5 )。结论 适当剂量 β射线局部照射可以抑制自体移植静脉的内膜增生及平滑肌细胞(SMC)的增殖 ,减少再狭窄的发生。
Objective To study the effect of radioisotope local irradiation on intimal hyperplasia in autologous vein graft and its possible mechanism. Methods A total of 40 autologous vein graft models were established and randomly divided into control group (autologous vein graft group), 8, 18, and 36 Gy groups (with different 32 P intimal irradiation doses), with 10 in each group. After 2 weeks, the specimens were fixed and stained with HE staining, Verhoeff elastic fiber staining and PCNA immunohistochemistry (SP method). The thickness of the intima of the transplanted vessels was measured by computer image analyzer. Results The mean intimal thickness of the vein graft was not significantly different between the control group and the 8Gy group (P> 0.05), but higher than that of the 18G and 36Gy groups (P <0 05). There was no significant difference between the 18Gy group and the 36Gy group (P> 0.05). PCNA positive cells showed that VSMC proliferation in 18,36 Gy group was significantly inhibited compared with that in control group (P <0.01). There was no significant difference between the two groups (P> 0.05). There was no significant difference between the 8Gy group and the control group (P> 0.05). Conclusion Local irradiation with appropriate dose of β-rays can inhibit intimal hyperplasia and proliferation of smooth muscle cells (SMC) in autologous vein and reduce the incidence of restenosis.
