目的 采用血清药理学的方法研究绿谷灵芝含药血清对氧化型低密度脂蛋白 (ox LDL)和糖化白蛋白诱导的内皮细胞表面血管细胞粘附分子 (VCAM 1)和细胞间粘附分子 (ICAM 1)表达的影响。方法 通过细胞ELISA和流式细胞仪技术检测细胞表面VCAM 1和ICAM 1表达的变化。结果 细胞ELISA结果显示 0 2 4 g·kg-1、0 72 g·kg-1绿谷灵芝连续灌胃给药 10d后取血分离得到的血清可降低糖化白蛋白诱导的内皮细胞表面VCAM 1和ICAM 1表达 (P <0 0 5 ) ,降低ox LDL诱导的ICAM 1表达 (P <0 0 5 ) ;流式细胞技术检测结果显示 0 72 g·kg-1绿谷灵芝含药血清可降低ox LDL和糖化白蛋白诱导的内皮细胞表面ICAM 1表达(P <0 0 5 ) ,0 12 g·kg-1绿谷灵芝含药血清和无药血清对内皮细胞表面粘附分子的表达无影响。结论 绿谷灵芝含药血清可降低糖化白蛋白和ox LDL诱导的内皮细胞表面粘附分子的表达
Objective To investigate the serum vascular endothelial cell adhesion molecule (VCAM 1) and intercellular adhesion molecule on vascular endothelial cells induced by oxidized low-density lipoprotein (ox-LDL) and glycated albumin using serum pharmacology. ICAM 1) The effect of expression. Methods The expression of VCAM 1 and ICAM 1 on the cell surface was detected by cell ELISA and flow cytometry. RESULTS: The results of cell ELISA showed that sera obtained from blood samples obtained after continuous intragastric administration of 0 24 g·kg-1 and 0 72 g·kg-1 Ganoderma lucidum for 10 days could reduce the glycosylated albumin-induced VCAM 1 on the endothelial cells. ICAM 1 expression (P <0 05) decreased ox-LDL-induced ICAM 1 expression (P <0 05); flow cytometry results showed that 0 72 g · kg-1 Lugu Ganoderma drug-containing serum reduced ox LDL and glycated albumin-induced endothelial ICAM 1 expression (P <0 05), 0 12 g · kg-1 Lugu Ganoderma drug-containing serum and drug-free serum had no effect on the expression of endothelial cell adhesion molecules. Conclusion Lugu Ganoderma lucidum serum can reduce the expression of endothelial cell adhesion molecules induced by glycated albumin and ox LDL.
