以忍冬属不同来源的4种种质为试材,采用形态分类学的方法,根据株高、叶长、叶宽、叶的形状、花开时间等农艺性状,对不同种质进行了比较分析,同时采用分子生物学的方法,以5对引物对4份种质的DNA条形码序列进行PCR扩增和测序,构建聚类图,并分析物种的亲缘关系。结果表明:忍冬属4种种质在农艺学性状上差异不明显,如采用形态学的方法进行鉴别容易导致误判,然而这4种种质在DNA条形码序列上存在着丰富的遗传多样性,研究认为在对金银花种质进行亲缘关系鉴定时,借助DNA条形码序列特性与农艺学性状相结合能有效区分物种之间的差异,鉴定结果相对可靠。
Four germplasms from different sources of Lonicera were selected as test materials. Morphological taxonomy was used to analyze the different germplasms based on the plant height, leaf length, leaf width, leaf shape, flowering time and other agronomic traits. At the same time, using molecular biology method, 5 pairs of primers were used to amplify and sequence DNA sequences of 4 germplasms. The cluster diagram was constructed and the genetic relationship of species was analyzed. The results showed that there was no significant difference in the agronomic traits between the four species of Lonicera. If the morphological methods were used to identify them, they could easily lead to miscarriage of justice. However, the genetic diversity of the four germplasms was rich in DNA barcode sequences. In the genetic relationship between honeysuckle germplasm identification, with the combination of DNA barcode sequence characteristics and agronomic traits can effectively distinguish between species differences, the identification results are relatively reliable.
