目的分析组织因子(TF)表达对人类大肠癌细胞侵袭和血行转移能力的影响。方法利用构建有正义/反义组织因子cDNA(TFcDNA)的质粒pcDNA3.1/Zeo,以脂质体法转染人类大肠癌细胞HT29细胞及LoVo细胞;转染成功的HT29细胞及LoVo细胞采用WesternBlot检测TF表达水平,通过裸鼠皮下成瘤观察细胞体内侵袭能力的变化;通过建立裸鼠体内肺转移及肝转移模型观察细胞体内血行转移能力的变化。结果转染了正义TFcDNA的HT29细胞及LoVo细胞的TF表达水平较未转染的HT29细胞及LoVo细胞升高,转染了反义TFcDNA的HT29细胞及LoVo细胞的TF表达水平则降低;转染了正义TFcDNA的HT29细胞的体内侵袭能力较未转染的HT29细胞增强,转染了反义TFcDNA的HT29细胞的体内侵袭能力则减弱;转染了正义TFcDNA的LoVo细胞的体内血行转移能力较未转染的LoVo细胞增强,转染了反义TFcDNA的LoVo细胞的体内血行转移能力能力则减弱。结论TF可以促进人类大肠癌细胞的侵袭和血行转移的能力。
Objective To analyze the effect of tissue factor (TF) on the invasion and hematogenous metastasis of human colorectal cancer cells. METHODS: Human colon cancer HT29 cells and LoVo cells were transfected with plasmid pcDNA3.1 / Zeo constructed by sense / antisense tissue factor cDNA (TFcDNA). The transfected HT29 cells and LoVo cells were transfected by Western Blot The expression level of TF was detected and the invasion ability of cells was observed by subcutaneous tumorigenesis in nude mice. The changes of hematogenous metastasis in vivo were observed by establishing lung metastasis and liver metastasis models in nude mice. Results The TF expression of HT29 cells and LoVo cells transfected with sense TF cDNAs were higher than those of untransfected HT29 cells and LoVo cells. The expression of TF in transfected HT29 cells and LoVo cells transfected with antisense TF cDNA decreased. The in vivo invasion ability of HT29 cells with sense TFcDNA was enhanced compared with that of untransfected HT29 cells, while the invasion ability of HT29 cells transfected with antisense TFcDNA was weakened. The ability of in vivo hematogenous metastasis of HT29 cells transfected with sense TFcDNA was less than that of HT29 cells transfected with sense TFcDNA The transfected LoVo cells were enhanced and the ability of LoVo cells transfected with antisense TF cDNA to reduce the hematogenous metastasis in vivo. Conclusion TF can promote the invasion and metastasis of human colorectal cancer cells.
