目的通过对胃癌组织中Evi-1及c-myb基因的检测,探讨2个基因在胃癌中的表达水平,及其与超声内镜下胃癌分期、病理分化程度的相关性。方法经内镜取活检,病理检查确诊分为低分化腺癌、中分化腺癌、高分化腺癌。超声内镜下胃癌浸润深度T分期为T1期、T2期、T3期、T4期,用实时定量荧光PCR法(Real-time)检测Evi-1及c-myb基因在不同病理分化程度及不同超声内镜分期癌组织、萎缩性胃炎组织及正常组织中的表达。结果 Evi-1基因在胃癌组织中高表达,在正常对照组及萎缩性胃炎中低表达。Evi-1基因的表达在胃癌组与正常对照组比较(P=0.00)、胃癌组与萎缩性胃炎组(P=0.018)比较差异均有显著性;Evi-1基因在转移组与非转移组比较(P=0.015)差异有显著性。Evi-1基因表达与病理分化程度(P=0.70)及超声内镜分期比较(P=0.46)差异无显著性。c-myb基因在3组均表达,胃癌组与正常对照组(P=0.29)及萎缩性胃炎组(P=0.50)比较差异无显著性。结论 Evi-1基因在胃癌组织中高表达,淋巴结转移组中该基因表达明显增高,但其表达与病理分化程度及超声內镜分期差异无显著性;Evi-1基因表达与胃癌转移有关,有望成为胃癌的诊断指标,并判断胃癌的预后,为进一步靶向治疗提供依据。
Objective To investigate the expression of Evi-1 and c-myb in gastric cancer and to investigate the correlation between the expression of Evi-1 and c-myb in gastric cancer and their relationship with the stage and pathological differentiation of gastric cancer under endoscopic ultrasonography. Methods Endoscopic biopsy, pathological examination diagnosed as poorly differentiated adenocarcinoma, moderately differentiated adenocarcinoma, well-differentiated adenocarcinoma. Endoscopic ultrasonography showed that the T stage of gastric cancer invasion was T1, T2, T3 and T4. Real-time quantitative RT-PCR was used to detect the expression of Evi-1 and c-myb in different pathological grades and different ultrasonography Endoscopic staging cancer tissue, atrophic gastritis and normal tissue expression. Results Evi-1 gene was highly expressed in gastric cancer tissues and was low in normal control group and atrophic gastritis. Evi-1 gene expression in gastric cancer group compared with the normal control group (P = 0.00), gastric cancer group and atrophic gastritis group (P = 0.018) were significantly different; Evi-1 gene in metastasis group and non-metastasis group The difference was significant (P = 0.015). Evi-1 gene expression and pathological differentiation (P = 0.70) and endoscopic ultrasonography (P = 0.46) no significant difference. The c-myb gene was expressed in all three groups. There was no significant difference between gastric cancer group and normal control group (P = 0.29) and atrophic gastritis group (P = 0.50). Conclusion Evi-1 gene is overexpressed in gastric cancer and the expression of Evi-1 gene is significantly increased in lymph node metastasis group, but the expression of Evi-1 gene is not significantly different from that of pathological differentiation and endoscopic stage. Evi-1 gene expression is associated with gastric cancer metastasis and is expected to become Gastric cancer diagnosis indicators, and to determine the prognosis of gastric cancer, to provide the basis for further targeted therapy.
