目的研究CD40及CD154(CD40L)在不同时间段糖尿病肾损害大鼠外周血单个核细胞和肾小管上皮细胞的表达及作用。方法雄性SD大鼠40只,随机分为对照组10只和实验组30只。实验组用链脲佐菌素(STZ)60mg/kg一次性腹腔注射SD大鼠,分别于1、4、12周将模型组与对照组大鼠处死,流式细胞术测定外周血单个核细胞表面CD40及CD154的表达;取肾脏组织,做病理切片及抗CD40免疫组织化学染色。结果 HE染色显示,与正常对照组相比,模型组有炎症细胞浸润,肾小管上皮细胞损伤和肾小球膨胀,基底膜增厚。注射链脲佐菌素1周后与对照组相比,CD40-CD154+在单个核细胞中的表达无差异。4、12周与对照组相比,CD40-CD154+在单个核细胞中的表达上升(P<0.05);免疫组化染色显示,4周和12周肾实质细胞CD40表达高于对照组(P<0.05)。结论在模型组外周血单个核细胞表面CD154的表达升高和肾小管上皮细胞CD40的表达上调在糖尿病大鼠的肾损伤中起了非常重要的作用。
Objective To study the expression of CD40 and CD154 (CD40L) on peripheral blood mononuclear cells and renal tubular epithelial cells in diabetic nephropathy rats at different time points. Methods Forty male SD rats were randomly divided into control group (n = 10) and experimental group (n = 30). In the experimental group, SD rats were injected intraperitoneally with streptozotocin (STZ) 60mg / kg, and the rats in the model group and the control group were sacrificed at 1, 4 and 12 weeks respectively. The levels of peripheral blood mononuclear cells Surface CD40 and CD154 expression; take kidney tissue, do pathological sections and anti-CD40 immunohistochemical staining. Results HE staining showed that compared with the normal control group, the model group had inflammatory cell infiltration, renal tubular epithelial cell injury and glomerular dilation, and thickening of the basement membrane. Compared with the control group, the expression of CD40-CD154 + in mononuclear cells after streptozotocin injection for 1 week showed no difference. The expression of CD40-CD154 + in mononuclear cells increased at 4 and 12 weeks (P <0.05). The expression of CD40 in renal parenchymal cells at 4 and 12 weeks was higher than that in control group (P < 0.05). Conclusion The increased expression of CD154 on peripheral blood mononuclear cells and the upregulation of CD40 expression in renal tubular epithelial cells play an important role in the renal injury of diabetic rats.
