目的:大鼠后脚掌注射完全弗氏佐剂(complete freund’s adjuvant,CFA)可诱导产生条件位置回避(conditioned place avoidance,C-CPA)反应,大鼠经过“非痛环境”/“痛环境”适应后,检测形成的C-CPA行为反应可持续多长时间。方法:(1)根据后脚掌注射CFA后观察痛行为的时间,将大鼠随机分为Day3组、Day5组、Day7组、Day10组、Day14组共五个实验组;(2)建立CFA诱导的CPA模型;(3)检测各组大鼠CPA反应;(4)Western-blot检测r ACC脑区痛情绪相关NMDA受体亚基NR1及磷酸化(p-NR1)的表达水平。结果:大鼠后脚掌注射CFA后五组大鼠热痛行为反应随时间无明显变化;CPA反应在Day3组、Day5组、Day7组、Day10组大鼠与注射CFA前相比产生了明显的CPA反应,而Day14组的CPA反应明显减弱;同步Western-blot检测的p-NR1表达也呈现了与行为反应一致的变化趋势。结论:CFA一次注射与环境条件化适应建立的C-CPA模型建成后可维持至少10 d的时间,之后CPA反应逐渐消失,且NMDA受体NR1亚基磷酸化水平与CPA反应的形成有关。
OBJECTIVE: Rats were injected with complete freund’s adjuvant (CFA) to induce conditioned place avoidance (C-CPA) response in rats. The rats were subjected to “pain-free” Environment "to determine how long the resulting C-CPA response can last. Methods: (1) According to the time of pain behavior after CFA injection, the rats were randomly divided into five groups: Day3 group, Day5 group, Day7 group, Day10 group and Day14 group. (2) CPA model; (3) CPA responses in each group were detected; (4) Western blot analysis was used to detect the expressions of NMDA receptor subunits NR1 and phosphorylation (p-NR1) in rAC brain regions. Results: There was no significant change in the thermal pain behavior of rats in the five groups after CFA injection into the rat paw. The CPA responses of rats in Day 3, Day 5, Day 7 and Day 10 groups were significantly higher than those before CFA injection Response, while the Day14 group CPA response was significantly weakened; simultaneous Western-blot detection of p-NR1 expression also showed a trend consistent with behavioral responses. CONCLUSION: The C-CPA model established by CFA injection and environmental conditioning can be maintained for at least 10 days after which the CPA response gradually disappears. The phosphorylation level of NR1 subunit of NMDA receptor is related to the formation of CPA reaction.
