目的通过检测大鼠全脑缺血后再灌流血浆及脑组织中内皮素的含量变化,初步探讨了内皮素与脑秧血再灌流后海马组织损伤的关系。方法利用四血管阻断法形成大鼠全脑缺血模型,在全脑缺血10min后再灌流6h、24h、72h以及120h后分别检测血浆及脑组织中内皮素的含量。结果大鼠全脑缺血再灌流后血浆及脑组织中内皮素含量明显升高,特别是海马组织中内皮素的增高最为明显。内皮素的增高在脑缺血后24-72h达到最高峰,120h有下降趋势。结论内皮素的改变与脑缺血再灌流引起的海马神经元迟发性坏死有密切的关系。
Objective To explore the relationship between endothelin and damage of hippocampus tissue after cerebral ischemia-reperfusion injury by detecting the content of endothelin in plasma and brain tissue after reperfusion in rats. Methods The rat model of global cerebral ischemia was established by four-vessel occlusion. Endothelins in plasma and brain tissue were detected at 6h, 24h, 72h and 120h after reperfusion 10 min after global cerebral ischemia. Results The content of endothelin in plasma and brain tissue was significantly increased after global cerebral ischemia and reperfusion in rats, especially the increase of endothelin in hippocampus. The increase of endothelin peaked at 24-72h after cerebral ischemia and decreased at 120h. Conclusion The change of endothelin is closely related to the delayed necrosis of hippocampal neurons induced by cerebral ischemia-reperfusion.
