目的:观察补阳还五汤、血府逐瘀汤对局灶性脑缺血大鼠海马脑源性神经营养因子(BDNF)蛋白表达的影响,探讨不同活血配伍方法在不同时期对BDNF含量的影响。方法:将大鼠随机分为假手术组、模型组、补气活血组、行气活血组,共4组,每组30只,采用局灶性脑缺血再灌注损伤大鼠模型,造模后给予正常组、模型组生理盐水灌胃10 m L·kg~(-1),补气活血组、行气活血组分别灌胃中药26 g生药·kg~(-1),14mg生药·kg~(-1),1次/d,分别在第5、10、15天断头处死,每次每组大鼠10只。光镜下观察海马病理学变化,免疫组化检测BDNF蛋白的表达。结果:补气活血组、行气活血组BDNF含量明显升高,与模型组比较,差异有统计学意义(P<0.01);补气活血组在两周之内呈现缓慢持续不断增高趋势,行气活血组在第5天达到峰值,之后呈下降趋势;在第10、15天,补气活血明显高于行气活血组(P<0.01)。结论:补气活血和行气活血的配伍均能升高脑组织BDNF的含量,但是补气活血作用强于行气活血,对脑组织具有更好的保护作用。
Objective: To observe the effect of Buyang Huanwu Decoction and Xuefu Zhuyu Decoction on the expression of brain-derived neurotrophic factor (BDNF) protein in hippocampus after focal cerebral ischemia in rats, and to explore the effect of different activating blood compatibility methods on BDNF content influences. Methods: The rats were randomly divided into sham-operated group, model group, qi-invigorating and blood-activating group, and qi-activating and blood-activating group. There were 4 rats in each group. The rats were subjected to focal cerebral ischemia-reperfusion injury The rats in model group were treated with normal saline (10 m L · kg ~ (-1)), Buqi Huoxue group and Xieqihuoxue group by intragastric administration of 26 g crude drug · kg -1, 14 mg crude drug · kg ~ (-1), once a day, and were decapitated on the 5th, 10th, and 15th day respectively, 10 rats in each group. The pathological changes of hippocampus were observed under light microscope, and the expression of BDNF protein was detected by immunohistochemistry. Results: Compared with the model group, the content of BDNF in qi and activating blood group and qi and activating blood group were significantly increased (P <0.01). The qi and blood activating group showed a slowly increasing trend in two weeks Qi-activating blood group reached its peak on the 5th day and then decreased. On the 10th and 15th days, Qi-activating blood circulation was significantly higher than that on the qi-activating blood circulation group (P <0.01). CONCLUSION: The combination of qi and activating blood circulation and qi and activating blood circulation can both increase the content of BDNF in brain tissue, but the effect of qi and activating blood circulation is stronger than that of qi and activating blood circulation and has a better protective effect on brain tissue.
