目的:研究雷公藤多苷(glycosides of Tripterygium wilfordii,GTW)对幼年雄性大鼠血清睾酮及睾丸组织中睾酮生成酶细胞色素P450胆固醇侧链裂解酶(cyto-chrome P450 side chain cleavage,P450scc)的影响以及补肾中药(菟丝子黄酮、六味地黄丸)对其损伤的干预作用。方法:3~4周龄SD雄性幼鼠48只,随机分为4组:空白组(CMC-Na)、多苷组(9 mg.kg-1.d-1)单独ig,六味组(六味地黄丸1 g.kg-1.d-1+GTW 9 mg.kg-1.d-1),黄酮组(菟丝子黄酮0.1 g.kg-1.d-1+GTW 9mg.kg-1.d-1)混合ig,持续给药12周后,处死大鼠留取血清和睾丸组织,采用放免法检测血清睾酮浓度;采用RT-PCR和Western blot检测睾丸组织中p450scc在基因和蛋白水平的表达。结果:GTW组与空白组相比可以明显降低血清睾酮(P<0.01);六味组与多苷组相比可以明显提高血清睾酮(P<0.05);在蛋白水平GTW与空白组相比可以明显降低睾丸组织P450scc(P<0.05);在基因水平GTW组与其他组比较无显著性差异。结论:GTW对幼年大鼠生殖系统有一定损伤作用;六味地黄丸可以通过提高雄鼠血清睾酮水平而达到保护生殖损伤的作用。
AIM: To investigate the effects of glycosides of Tripterygium wilfordii (GTW) on serum testosterone and cytochrome P450 side chain cleavage (P450scc) of testosterone-producing enzyme in young male rats As well as kidney medicine (dodder flavonoids, Liu Wei Di Huang Wan) on its injury intervention. Methods: Forty-eight SD male rats aged 3 to 4 weeks were randomly divided into 4 groups: control group (CMC-Na), polyglucoside group (9 mg.kg-1.d- Rehmannia glutinosa 1 g.kg-1.d-1 + GTW 9 mg.kg-1.d-1), flavonoids group (dodder flavone 0.1 g.kg-1.d-1 + GTW 9 mg.kg-1.d -1) were mixed ig. After 12 weeks of continuous administration, serum and testicular tissue were collected from rats and serum testosterone concentration was determined by radioimmunoassay. The expression of p450scc in testis tissues was detected by RT-PCR and Western blot . Results: Compared with blank group, GTW group could significantly reduce serum testosterone (P <0.01); Liuwei group could significantly increase serum testosterone compared with polyglucoside group (P <0.05); GTW protein group was significantly lower than blank group Reduce testicular P450scc (P <0.05); there was no significant difference between GTW group and other groups at gene level. CONCLUSION: GTW can damage the reproductive system of juvenile rats. Liuweidihuang Pills can protect reproductive injury by increasing serum testosterone levels in male rats.
