探讨经结核杆菌抗原特异性活化的γδT细胞的抗肿瘤活性。方法 :结核杆菌抗原 (Mtb Ag)、IL 2、PHA做为刺激原 ,活化正常人外周血单个核细胞 (PBMC) ,用流式细胞仪分析活化后淋巴细胞的表型 ,用MTT法测定活化淋巴细胞的杀伤活性。结果 :Mtb体外活化PBMC可于 18天获得大量淋巴细胞 ,且γδT细胞占有较高比例 ,对肿瘤细胞有较高杀伤活性。结论 :用Mtb为刺激原可获得大量γδT细胞 ,且在体外对肿瘤细胞有较高的杀伤活性。
To investigate the anti-tumor activity of γδT cells activated by Mycobacterium tuberculosis antigen. Methods: Peripheral blood mononuclear cells (PBMCs) were activated by Mtb Ag, IL2 and PHA. The phenotypes of activated lymphocytes were analyzed by flow cytometry and activated by MTT assay Lymphocyte killing activity. Results: Mtb activation of PBMC in vitro in 18 days to obtain a large number of lymphocytes, and γδ T cells occupy a higher proportion of the tumor cells have higher killing activity. CONCLUSION: A large number of γδT cells can be obtained by using Mtb as stimulator, and have high cytotoxic activity against tumor cells in vitro.
