目的:克服亲和层析纯化抗人绒毛膜促性腺激素抗体(抗hCG抗体)IgG中载体与配体联接时不良的空间效应,以提高亲和层析的效率。方法:用6-氨基己酸为手臂通过溴化氰连接到Sepharose4B上形成一种新的层析介质,然后将hCG与之偶联用以纯化抗hCG的抗体IgG。结果:用该种亲和层析介质纯化的抗hCG抗体IgG回收率及免疫滴度均高于无手臂的亲和层析介质(活性回收率平均增加22.5%,比活性平均增加55.6%)。结论:用接手臂层析介质纯化抗hCG抗体IgG是一种比较好的方法。
OBJECTIVE: To overcome the adverse spatial effect of affinity chromatography of carrier and ligand in affinity chromatography purified anti-human chorionic gonadotropin (IgG) antibody IgG to improve the efficiency of affinity chromatography. METHODS: A new chromatographic medium was formed by attaching 6-aminocaproic acid as an arm to cyano-cyanogen bromide via cyanogen bromide and then coupled with hCG to purify IgG against hCG. Results: The IgG recoveries and immunoteric titres of anti-hCG antibodies purified by this affinity chromatography medium were higher than those without arm affinity chromatography medium (the average activity recovery rate increased by 22.5% and the specific activity increased by 55%). 6%). Conclusion: It is a good method to purify anti-hCG antibody IgG using arm chromatography medium.
