目的 探讨内皮素 (Endothelin ET- 1)在大肠腺癌血管形成中的作用。方法 应用免疫组织化学 SABC法 ,对 41例大肠腺癌和 2 0例腺瘤组织中 ET- 1,VEGF和 F RAg分别进行免疫特异性染色 ,并计数肿瘤组织内微血管密度 (MVD)。结果 ET- 1染色主要分布于肿瘤细胞胞浆 ,阳性肿瘤基质中血管内皮细胞及成纤维细胞也有较强的着色。腺癌中 ET- 1阳性表达率为 73.4% ,显著高于腺瘤的 45% (P<0 .0 5 )。等级相关分析表明 ,ET- 1与 VEGF及 MVD密切相关。 (rs’=0 .6 7,rs’=0 .5 9,P<0 .0 1)。值得注意的是 ,在 ET- 1和 VEGF均有表达的 2 6例大肠腺癌中 ,连续切片显示 VEGF表达阳性的区域 ,ET- 1也倾向于阳性 ,并且两者在血管内皮细胞均有表达。。结论 ET- 1参与大肠腺癌肿瘤血管形成 ,并且与 VEGF具有协同作用
Objective To investigate the role of Endothelin ET-1 in angiogenesis of colorectal adenocarcinoma. Methods Immunohistochemical SABC method was used to immunostaining ET-1, VEGF and F RAg respectively in 41 cases of colorectal adenocarcinoma and 20 cases of adenoma and counting the microvessel density (MVD) in the tumor tissue. Results ET-1 staining was mainly located in the cytoplasm of tumor cells, and the staining of endothelial cells and fibroblasts in the positive tumor matrix was also strong. The positive rate of ET-1 in adenocarcinoma was 73.4%, which was significantly higher than that of adenoma (P <0.05). Hierarchical correlation analysis showed that ET-1 is closely related to VEGF and MVD. (rs ’= 0 .6 7, rs’ = 0 .59, P <0 .01). It is worth noting that in the 26 cases of colorectal adenocarcinoma with both ET-1 and VEGF expression, serial sections showed positive VEGF expression, ET-1 also tended to be positive, and both were expressed in vascular endothelial cells . . Conclusion ET-1 is involved in angiogenesis of colorectal adenocarcinoma and has a synergistic effect with VEGF
