分别用酚 -硫酸法、L owry- Folin法和高效液相凝胶渗透色谱法 (HPGPC)测定云芝菌丝体提取物云芝糖肽 (1)中糖和蛋白质的含量及分子量。来自 8株菌株的提取物其多糖含量为 41.1%~ 47.4% (P>0 .0 5 ) ;蛋白质含量为2 3.1%~ 36 .6 % (P<0 .0 5 ) ,分子量为 34k D~ 6 8k D。云芝菌丝体经碱提得到的提取物 1- K,多糖含量和分子量分别为 11.0 %和 14k D,均低于其进一步的醇沉物 1- KA(35 .8%和 38k D) ,但两者蛋白质含量为 36 .4%和 38.5 % ,无显著差异。菌丝体水提物 1- U A经纯化后得 1- U C,其多糖含量和分子量分别上升到 71.4%和 5 3k D。
The content of carbohydrate and protein and the molecular weight of the polysaccharide of Gemcitabine (1) were determined by phenol-sulfuric acid method, L owry-Folin method and high performance liquid chromatography gel permeation chromatography (HPGPC). The polysaccharide content of the extracts from 8 strains was 41.1% ~ 47.4% (P> 0.05), the protein content was 21.3% ~ 36.6% (P <0.05), the molecular weight was 34k ~ 6 8k D. The polysaccharide content and molecular weight of the extract obtained by alkali extraction were 11.0% and 14k D, respectively, which were lower than those of the further alcohol precipitates 1-KA (35.8% and 38k D) However, the protein content of both was 36.4% and 38.5%, no significant difference. The mycelium water extract 1-U A was purified to give 1-U C, its polysaccharide content and molecular weight increased to 71.4% and 53kD, respectively.
