目的:通过观察胃癌细胞核转录因子NF-κBP65蛋白与增殖细胞核抗原(PCNA)蛋白的表达及关系,探讨消痰散结方抑制胃癌细胞增殖的作用机制。方法:采用OB胶粘贴法建立裸鼠人胃癌MKN-45原位移植瘤模型,随机数字表法分成模型组,消痰散结低、中、高剂量组,5-Fu组,每组8只,观察消痰散结方干预对胃癌组织核转录因子NF-κBP65蛋白、增殖细胞核抗原(PCNA)表达的影响及相关性。结果:①消痰散结方中、高剂量组、5-Fu组NF-κB的P65蛋白的阳性表达与模型组相比P<0.05,有统计学意义;②消痰散结方低、中、高剂量组、5-Fu组的PCNA阳性表达率与模型组相比有统计学意义,P<0.05;③胃癌细胞NF-κBP65蛋白与PCNA表达存在正相关(r=0.857,P<0.01)。结论:胃癌MKN-45细胞中存在着活化的NF-κB及高表达的PCNA,并且存在线性相关,消痰散结方可能通过其抑制NF-κB活性,阻断了NF-κB对PCNA的活性调控,使胃癌细胞增殖受到抑制,控制胃癌复发与转移。
OBJECTIVE: To investigate the mechanism of Xiao Tan San Jie Fang inhibiting the proliferation of gastric cancer cells by observing the expression of NF-κB P65 and PCNA in gastric cancer cells. Methods: The nude mice gastric cancer MKN-45 orthotopically transplanted tumor model was established by OB adhesive method. The mice were randomly divided into model group, Xiaotan Sanjie low, medium and high dose group, 5-Fu group, 8 Only observe the influence of Xiaotan Sanjie Recipe on the expression of nuclear transcription factor NF-κBp65 and PCNA in gastric cancer tissues and their correlation. Results: ①The positive expression of P65 protein in NF-κB in middle-dose and high-dose group of Xiaotan Sanjie prescription group was significantly lower than that in model group (P <0.05); ② Xiaotan Sanjie prescription was low, , High-dose group and 5-Fu group were significantly higher than those in model group (P <0.05). ③ There was a positive correlation between NF-κBp65 protein and PCNA expression in gastric cancer cells (r = 0.857, P <0.01) . Conclusion: MKN-45 cells in gastric cancer exist active NF-κB and high expression of PCNA, and there is a linear correlation, Xiaotan Sanjie may inhibit NF-κB activity, blocking the activity of NF-κB on PCNA Regulated, so that inhibition of gastric cancer cell proliferation, control of gastric cancer recurrence and metastasis.
