目的 :建立肝复康丸中五味子甲素和五味子乙素的含量测定方法。方法 :样品用己烷超声提取 ,提取液过滤 ,蒸干。残渣用甲醇溶解后 ,注入高效液相色谱仪分离测定。色谱条件包括 :LUNA -C1 8分析色谱柱 ,柱温 30℃。乙腈 -水 (6 8∶32 )混合液(含 0 .2 %磷酸 ,用三乙胺调pH5 )作为流动相。检测波长为 2 5 4nm。结果 :肝复康丸中五味子甲素和五味子乙素与其它成份分离良好。五味子甲素峰和五味子乙素峰保留时间分别约为 16min和 2 1min。五味子甲素和五味子乙素峰与其相邻峰的分离度大于 1.5。以五味子甲素峰计算理论板数是 6 0 70。以五味子乙素峰计算理论板数是 70 15。五味子甲素对照品线性浓度范围35 .0 0~ 140 .0 μg·ml- 1 ,r =0 .9999,平均回收率为 10 0 .5 % ,RSD为 1.5 % (n =5 )。五味子乙素对照品的线性浓度范围 2 5 .0 0~ 10 0 .0 μg·ml- 1 ,r =0 .9999,平均回收率为 10 2 .4% ,RSD为 1.7% (n =5 )。结论 :本法灵敏度高、操作简便、结果准确 ,可用于肝复康丸中五味子甲素和五味子乙素的含量测定。
Objective : To establish a method for the determination of schisandrin and schisandrin B in Ganfukang Pills. Methods: The samples were extracted with hexane and the extracts were filtered and evaporated to dryness. The residue was dissolved in methanol and injected into a high performance liquid chromatograph for separation and measurement. Chromatographic conditions include: LUNA-C18 analysis column, column temperature 30 °C. Acetonitrile-water (6 8:32) mixture (containing 0.2% phosphoric acid, adjusted to pH 5 with triethylamine) was used as the mobile phase. The detection wavelength is 254 nm. Results : The schisandrin A and schisandrin B in Ganfukang Pill were separated well from other ingredients. The retention time of schisandra A and schisandra B peaks were approximately 16 min and 2 1 min, respectively. The separation of schizandrin and schisandrin B peaks from their adjacent peaks is greater than 1.5. The theoretical plate number for Schisandra A peak is 6070. The number of theoretical plates calculated with Schizandrin B peaks is 70 15 . The linear concentration range of schisandrin A reference substance was 35.0 to 140.0 μg·ml-1, r=0.9999, the average recovery was 10 0.5%, and the RSD was 1.5% (n =5). The linear concentration range of schisandrin B reference substance was 25.0 to 1.00.0 μg·ml-1, r=0.9999, the average recovery was 102.4%, and the RSD was 1.7% (n=5). . Conclusion: This method has high sensitivity, simple operation and accurate results. It can be used for the determination of schisandrin and schisandrin B in Ganfukang Pill.
