第3号染色体短臂(3p)的杂合子丢失(LOH)在很多肿瘤中已有报道。作者过去研究证实,50%口腔癌3p上有3个不同缺失区域(3p13-21.1,3p21.3-23和3p 25)。本研究检查了30例口腔异常增生(DLs)3pLOH,验证3号染色体畸变是否是口腔癌发生的早期信号。 30例口腔DLs来自27名患者,临床表现为白斑和红斑,组织学表现为重度异常增生(SED)或原位癌(CIS),以正常粘膜为对照。提取基因组DNA,用15种3p和2种3q微卫星标志为引物,PCR扩增后产物进行分析。结果:①3p等位基因不平衡频率:43%口腔DLs存在一个或多个等位基因不平衡,其中1例3个不连续区域,9例2个不连续区域,3例单个区域等位
Loss of heterozygosity (LOH) on the short arm of chromosome 3 (3p) has been reported in many tumors. In the past, the authors confirmed that there are three different regions of loss in 3% of the 50% oral carcinomas (3p13-21.1, 3p21.3-23 and 3p25). This study examined 30 cases of oral dysplasia (DLs) 3pLOH to verify whether chromosome 3 aberration is an early sign of oral carcinogenesis. Thirty oral DLs were obtained from 27 patients with clinical manifestations of leukoplakia and erythema, histological findings of severe dysplasia (SED) or carcinoma in situ (CIS), and normal mucosa as a control. Genomic DNA was extracted and analyzed by PCR using 15 3p and 2 3q microsatellite markers as primers. RESULTS: ①3p allele imbalance frequency: 43% of oral DLs had one or more allele imbalances, including 1 case of 3 discontinuous regions, 9 cases of 2 discontinuous regions and 3 cases of single region alleles
