嘌呤在肿瘤的发展过程中扮演着重要的角色,该信号通路受到一系列核苷酸酶调控。在这些酶中,CD73是关键酶之一,可以催化磷酸腺苷(AMP)水解为腺苷,从而介导腺苷的生物学作用。目前的研究已经发现CD73高表达于多种实体肿瘤,多个因子和机制都参与CD73表达的调控。越来越多的研究发现,CD73是体外肿瘤细胞增殖、转移和侵袭,体内肿瘤血管生成和肿瘤免疫逃逸机制的关键调控分子。体内试验的证据显示,对于肿瘤患者,靶向阻断CD73将成为有效的治疗手段。文章总结了CD73在肿瘤发展过程中的重要作用,包括其对于肿瘤血管生成的促进作用、影响肿瘤生长和转移的机制、对肿瘤微环境中免疫系统的抑制效应以及未来抗CD73分子的治疗与展望。
Purines play an important role in the development of tumors, which are regulated by a series of nucleotidases. Of these enzymes, CD73 is one of the key enzymes that catalyzes the hydrolysis of adenosine monophosphate (AMP) to adenosine thereby mediating the biological effects of adenosine. The current study has found that CD73 is highly expressed in many solid tumors, and many factors and mechanisms are involved in the regulation of CD73 expression. More and more studies have found that CD73 is a key regulator of in vitro tumor cell proliferation, metastasis and invasion, in vivo tumor angiogenesis and tumor immune escape mechanism. Evidence from in vivo studies has shown that targeted blocking of CD73 will be an effective treatment for cancer patients. The article summarizes the important role of CD73 in tumor development, including its promoting effect on tumor angiogenesis, the mechanism of affecting tumor growth and metastasis, the inhibitory effect on the immune system in the tumor microenvironment, and the future treatment and prospect of anti-CD73 molecule .
