目的:研究人骨肉瘤组织中Ⅰ、Ⅱ和Ⅲ型胶原的表达及其与骨肉瘤分型和分化的关系,并对胶原的两种染色方法进行比较。方法:用饱和苦k味酸.天狼星红-偏振光镜和免疫组化LSAN法,检测46例人骨肉瘤组织中Ⅰ、Ⅱ和Ⅲ型胶原的表达情况,并用EMAL-200真彩图像分析系统进行分析和半定量测定。结果:骨肉瘤组织中不仅有Ⅰ型胶原的表达(阳性率87%,灰度值171.99±14.74),且出现了Ⅱ型和Ⅲ型胶原(阳性率分别为30.4%和43.4%,灰度值分别为153.071±18.82和168.29±18.36);与对照的骨瘤组织比较,骨肉瘤Ⅰ型胶原表达明显减弱;Ⅱ型胶原在软骨母细胞型和混合型的表达明显高于其它型骨肉瘤(P<0.of),而Ⅰ、Ⅲ型胶原的表达在各型骨肉瘤间差异无显著性;在高中低3组不同分化程度的骨肉瘤间,Ⅰ型胶原表达差异有显著性(P<001),分化越低表达越少;天狼星红与免疫组化染色结果基本一致。结论:Ⅱ型胶原可作为软骨母细胞型和混合型骨肉瘤的分型参考指标,Ⅰ型胶原的表达可作为骨肉瘤分化及恶性度判断的参考指标。天狼星红染色是鉴别Ⅰ、Ⅲ型胶原简便、经济的方法之一。
Objective: To study the expression of type I, II and III collagen in human osteosarcoma and its relationship with osteosarcoma typing and differentiation, and to compare the two collagen staining methods. Method: Saturated bitter k-acid. Sirius red-polarized light microscopy and immunohistochemical LSAN method were used to detect the expression of type I, II and III collagen in 46 cases of human osteosarcoma, and analyzed by EMAL-200 true color image analysis system and semi-quantitatively measured. RESULTS: In osteosarcoma, there was not only the expression of type I collagen (positive rate 87%, gray value 171.99±14.74), but also type II and type III collagen (positive rate was 30.4% and 43.4%, gray values were 153.071±18.82 and 168.29±18.36, respectively; compared with the control osteosarcoma tissues, the expression of type I collagen was significantly decreased in osteosarcoma; type II collagen was in the cartilage mother. The expression of cell type and mixed type was significantly higher than that of other types of osteosarcoma (P<0.of), but the expression of type I and type III collagen was not significantly different among various types of osteosarcoma; in the high, middle and low groups, the degree of differentiation was different. Among the osteosarcomas, there was a significant difference in the expression of type I collagen (P < 001). The lower the differentiation, the lower the expression. The results of Sirius erythematosus and immunohistochemical staining were basically the same. Conclusion: Type II collagen can be used as a reference marker for chondroblastoma and mixed osteosarcoma, and the expression of type I collagen can be used as a reference index for osteosarcoma differentiation and malignancy judgment. Sirius red staining is one of the simple and economical methods to identify Type I and Type III collagen.
