本文报道了采用不同寄主小鼠进行小鼠造血干细胞体内扩散盒琼脂培养的结果比较。寄主小鼠在植入扩散盒1~2小时前接受750拉德~(60)Co照射或在植入前24小时注射VCR1.6毫克/公斤并未能提高CFU-C细胞团产率。这种情况是否是由于正常小鼠体液内的CSF水平已足以有效地刺激小鼠骨髓CFU-C生成细胞团,故提高CSF浓度不能再增加细胞团的产率。有待在掌握本实验小鼠的种属特性及调整照射剂量后作进一步研究。
In this paper, the results of in vivo diffusion kit agar culture of mouse hematopoietic stem cells using different host mice were compared. Host mice receiving 750 rad(60)Co 1 to 2 hours prior to implantation of the diffusion cassette or VCR 1.6 mg/kg 24 hours prior to implantation did not increase the CFU-C cell pellet yield. Whether this situation is due to the CSF level in the body fluid of normal mice is sufficient to effectively stimulate the mouse bone marrow CFU-C to generate cell mass, so increasing the concentration of CSF can no longer increase the yield of cell mass. To be further studied after mastering the species characteristics of the mice in this experiment and adjusting the irradiation dose.
