目的 :探讨分析金钗石斛多糖对2型糖尿病大鼠心功能、炎症因子以及心脏组织中RIP3的表达特点,以为金叉石斛多糖的开发和2型糖尿病心肌损伤的治疗提供参考。方法 :60只SD大鼠随机分为正常组、模型组、金钗石斛多糖低、中和高剂量干预组以及阳性药物贝那普利治疗组,每组10只,除正常组外,其余5组给予高脂高糖饮食6个月构建2型糖尿病大鼠模型,模型建立成功后,正常组和模型组给予1 m L·kg~(-1)·d~(-1)生理盐水灌胃,金钗石斛多糖3个干预组分别给予金钗石斛多糖100、200、400 mg·kg~(-1)·d~(-1)治疗,贝那普利治疗组则给予贝那普利1 mg·kg~(-1)·d~(-1)灌胃治疗,治疗8周后处死大鼠,HE染色检测肝脏病理学,试剂盒检测心功能和炎性因子,实时定量RT-PCR和Western blot检测RIP3 m RNA和蛋白的表达水平。结果 :正常组大鼠毛色光滑,活泼好动,模型组毛色发黄,同时精神萎靡,金钗石斛多糖3个干预组以及贝那普利治疗组相较于模型组有一定好转;HE染色可见正常组心肌细胞排列整齐,而模型组心肌细胞明显肥大,同时肌纤维排列紊乱和断裂;金钗石斛多糖干预后,其心肌细胞排列明显好转,特别是高剂量组更为明显;与正常组相比,模型组血糖、CK、CK-MB、LDH、IL-1α、IL-1β、IL-6和TNF-α和胰岛素抵抗指数明显升高(P<0.05),胰岛素含量明显降低(P<0.05),金钗石斛多糖3个干预组和贝那普利治疗组相较于模型组血糖、CK、CK-MB、LDH、IL-1α、IL-1β、IL-6和TNF-α和胰岛素抵抗指数均明显降低(P<0.05),胰岛素含量则明显升高(P<0.05),而贝那普利治疗组变化趋势基本介于金钗石斛多糖低剂量组和中剂量组之间;与正常组相比,模型组RIP3表达明显升高(P<0.05),而金钗石斛多糖和贝那普利均可以降低其RIP3表达(P<0.05)。结论 :金钗石斛多糖可改善2型糖尿病心肌损伤大鼠心功能和炎症反应,此外对心肌细胞凋亡也有一定的改善作用;这可能与金钗石斛多糖可调节糖尿病大鼠心肌组织中RIP3表达有关。
Objective: To investigate the characteristics of heart function, inflammatory factors and expression of RIP3 in heart tissue of type 2 diabetic rats induced by Dendrobium nobile polysaccharide, in order to provide reference for the development of dendrobium fimbriatum and the treatment of type 2 diabetes mellitus. Methods: Sixty Sprague-Dawley rats were randomly divided into normal group, model group, low, middle and high dose intervention group of dendrobium nobile dendritic cells and the positive drug benazepril treatment group, with 10 rats in each group. Except normal group, the other 5 The rats in model group were given high fat and high sugar diet for 6 months to establish type 2 diabetic rat model. After the model was established successfully, normal saline and normal saline were given to the model group by intragastric administration of 1 m L · kg -1 d -1 saline , And three polysaccharides from Dendrobium nobile were respectively treated with 100, 200 and 400 mg · kg -1 · d -1 polysaccharides from Dendrobium nobile, and benazepril-treated group kg · d -1 · d -1. After 8 weeks of treatment, the rats were killed, the pathology of liver was detected by HE staining, the cardiac function and inflammatory cytokines were detected by kit, the real-time quantitative RT-PCR and Western blot was used to detect the expression of RIP3 mRNA and protein. Results: In the normal group, the coat color was smooth and lively. The model group had yellow hair color and apathetic. The three intervention groups of dendrobium nobile and the benazepril treatment group were better than the model group. HE staining showed Normal group of myocardial cells arranged in neat rows, while the model group of myocardial cells significantly hypertrophy, while muscle fiber disorder and fracture arrangement; Dendrobium nobile polysaccharide intervention significantly improved myocardial cell arrangement, especially in the high-dose group was more obvious; compared with the normal group (P <0.05), and the levels of insulin, insulin and CK-CK, CK-MB, LDH, IL-1α, IL-1β, IL-6 and TNF-α in the model group were significantly increased , 3 groups of dendrobium nobile polysaccharides and benazepril treatment group compared with the model group, blood glucose, CK, CK-MB, LDH, IL-1α, IL-1β, IL-6 and TNF-α and insulin resistance index (P <0.05), while the content of insulin was significantly increased (P <0.05), while the change tendency of benazepril treatment group was basically between the low dose group and the middle dose group of Dendrobium nobile. Compared with the normal group Compared with model group, the expression of RIP3 was significantly increased (P <0.05), while the polysaccharide of Dendrobium nobile and benazepril decreased RIP3 expression (P <0.05). Conclusion: Dendrobium nobile polysaccharides can improve cardiac function and inflammatory response in myocardial tissue of type 2 diabetic rats, and also improve cardiomyocyte apoptosis. This may be related to that the polysaccharide from Dendrobium nobile can regulate the expression of RIP3 in diabetic rat heart related.
