目的:研究甘露糖受体在阴道上皮细胞的表达,以了解具有甘露糖残基的假丝酵母菌与阴道间的共生关系。方法:采用免疫组化研究甘露糖受体在人阴道上皮细胞的表达。免疫印迹研究人阴道上皮层甘露糖受体的表达,比较绝经组和未绝经组甘露糖受体的表达水平。结果:(1)巨噬细胞甘露糖受体多克隆抗体免疫组化显示甘露糖受体于阴道上皮细胞膜表达,同时阴道上皮固有层有阳性表达。而单克隆抗体免疫组化显示该受体仅于固有层表达;(2)免疫印迹结果示,在分子量约190kD处出现阳性表达条带;(3)未绝经组甘露糖受体相对表达量为0.86±0.45,高于绝经组0.46±0.23,差异显著(P<0.05)。结论:阴道上皮细胞膜表达一种甘露糖受体,它与巨噬细胞甘露糖受体分子结构可能不完全相同。假丝酵母菌黏附于阴道上皮细胞,可能是通过其甘露糖残基与阴道上皮细胞膜上的甘露糖受体识别而实现的。绝经组甘露糖受体表达下降,致使阴道上皮细胞与假丝酵母菌的黏附减少,假丝酵母菌失去致病的前提。
OBJECTIVE: To study the expression of mannose receptor in vaginal epithelial cells in order to understand the symbiotic relationship between Candida albicans and vagina. Methods: The expression of mannose receptor in human vaginal epithelial cells was studied by immunohistochemistry. Western blotting was used to investigate the expression of mannose receptor in human vaginal epithelium and to compare the expression of mannose receptor in menopausal and non-menopausal groups. Results: (1) The macrophage mannose receptor polyclonal antibody immunohistochemistry showed mannose receptor expression in the vaginal epithelial cell membrane, while the lamina propria positive expression. Immunohistochemistry showed that the receptor was only expressed in the lamina propria. (2) Immunoblotting showed that a positive expression band appeared at a molecular weight of about 190 kD. (3) The relative expression of mannose receptor in non-menopausal group was 0.86 ± 0.45, which was significantly higher than that of the menopause group (0.46 ± 0.23, P <0.05). Conclusion: The vaginal epithelial cell membrane expresses a mannose receptor, which may not be exactly the same as the macromolecular mannose receptor molecular structure. Candida adherence to vaginal epithelial cells may be through its mannose residues and vaginal epithelial cell membrane mannose receptor recognition achieved. Menopausal group mannose receptor expression decreased, resulting in decreased adhesion of vaginal epithelial cells and Candida, Candida loses the pathogenic premise.
