目的 研究三氧化二砷(As2O3)对人胃癌细胞株癌相关基因的影响,探讨其抗肿瘤作用。方法 以As2O3作用于体外培养的人胃癌细胞株,通过免疫组化技术对P53、Cyclin-D、PCNA基因编码蛋白的表达进行检测。结果 经As2O3作用的人胃癌细胞P53、Cyclin-D、PCNA基因编码蛋白表达减少。结论 初步证实As2O3具有下调P53、Cyclin-D、PCNA基因编码蛋白表达,抑制肿瘤的增殖和转移,从而具有抗肿瘤作用。
Objective To study the effect of arsenic trioxide (As2O3) on cancer-associated genes in human gastric cancer cell lines and to explore its anti-tumor effect. Methods As2O3 was used to in vitro cultured human gastric cancer cell lines, and the expressions of P53, Cyclin-D and PCNA genes were detected by immunohistochemistry. Results The expression of P53, Cyclin-D and PCNA genes in human gastric cancer cells treated with As2O3 decreased. Conclusion As2O3 has been shown to have the anti-tumor effect by down-regulating the protein expression of P53, Cyclin-D and PCNA genes and inhibiting the proliferation and metastasis of tumor cells.
