甜樱桃杂交胚组培适宜灭菌方法的研究

以甜樱桃杂交胚为试验材料,研究了不同灭菌组合及二次灭菌对杂交胚的灭菌效果,以期筛选出适合甜樱桃杂交胚组培的最佳灭菌方法。结果表明,甜樱桃杂交胚组培以使用75%酒精1min、10%次氯酸钠灭菌15min、0.1%升汞灭菌1min,用无菌水冲洗5次,用无菌滤纸吸干种胚表面水分,剥去种皮,再使用0.05%升汞灭菌0.5min后污染率、褐化率最低。
