为探讨氧化型低密度脂蛋白在血栓形成过程中的作用及机制,用超速离心法从正常人血浆中分离出低密度脂蛋白后,用Cu2 + 使之氧化变成氧化型低密度脂蛋白。在人脐静脉内皮细胞的培养基中加入一定浓度的低密度脂蛋白或氧化型低密度脂蛋白,培养一定时间后,用异硫氰酸胍法抽提细胞总RNA,用反转录聚合酶链反应方法检测人脐静脉内皮细胞中组织因子通路抑制子mRNA的表达,以观察氧化型低密度脂蛋白对人脐静脉内皮细胞表达组织因子通路抑制子mRNA 的影响。结果发现,人脐静脉内皮细胞能持续性地表达组织因子通路抑制子mRNA,它的表达不受低密度脂蛋白的影响。但氧化型低密度脂蛋白能明显地以时间和浓度依赖性方式抑制人脐静脉内皮细胞表达组织因子通路抑制子mRNA。实验结果提示氧化型低密度脂蛋白可能通过抑制血管内皮细胞表达组织因子通路抑制子,从而在诱导动脉粥样硬化斑块内血栓形成过程中发挥极为重要的作用。
To explore the role and mechanism of oxidized low density lipoprotein in thrombus formation, low density lipoproteins were isolated from normal human plasma by ultracentrifugation and oxidized to oxidized low density lipoprotein by Cu2 +. A certain concentration of low density lipoprotein or oxidized low density lipoprotein was added into the culture medium of human umbilical vein endothelial cells. After a certain period of culture, the total RNA was extracted by guanidine isothiocyanate method, Chain reaction method to detect the expression of tissue factor pathway inhibitor mRNA in human umbilical vein endothelial cells in order to observe the effect of oxidized low density lipoprotein on the expression of tissue factor pathway inhibitor mRNA in human umbilical vein endothelial cells. The results showed that human umbilical vein endothelial cells can persist expression of tissue factor pathway inhibitor mRNA, its expression from the impact of low density lipoprotein. However, oxidized low density lipoprotein significantly inhibited the expression of tissue factor pathway inhibitor mRNA in human umbilical vein endothelial cells in a time and concentration dependent manner. The experimental results suggest that oxidized low density lipoprotein may play an important role in inducing thrombosis in atherosclerotic plaques by inhibiting the expression of tissue factor pathway suppressors in vascular endothelial cells.
