目的:对牡丹皮中Fe,Mn,Cu,Zn,Al,Pb,As,Cd等24种微量元素采用煎煮法进行提取。方法:使用0.45μm的微孔滤膜分离提取液中的悬浮态和可溶态,将可溶态通过大孔树脂(LSA-10),以去离水和甲醇洗脱,确定微量元素的无机态和有机态;模拟人体胃肠溶液,采用正辛醇-水系统分离微量元素的醇溶态和水溶态;通过ICP-MS技术(电感耦合等离子体质谱法),测定微量元素总量及各种形态。结果:方法的回收率为74.7%~118.2%,精密度RSD在1.3%~6.9%。24种微量元素的溶出率为2.0%~82.9%,可溶态在水中的比率为73.6%~99.5%,可溶态中80%以上微量元素以无机态和水溶态形式存在。结论:微量元素的形态分析及溶出特征为牡丹皮的临床应用提供理论依据。
Objective: To extract 24 kinds of trace elements such as Fe, Mn, Cu, Zn, Al, Pb, As and Cd from Cortex Moutan by decoction method. Methods: The suspended state and soluble state in the extract were separated using a 0.45 μm microporous membrane. The soluble fraction was passed through a macroporous resin (LSA-10) and eluted with water and methanol to determine the trace inorganic element State and organic state. The human gastrointestinal solution was simulated. The n-octanol-water system was used to separate the alcohol-soluble and water-soluble trace elements. The content of trace elements and the contents of trace elements were determined by inductively coupled plasma mass spectrometry (ICP- Species. Results: The recoveries of the method were 74.7% ~ 118.2%, and the RSDs of precision were 1.3% ~ 6.9%. The elution rates of 24 kinds of trace elements ranged from 2.0% to 82.9%, the soluble ratios in water ranged from 73.6% to 99.5%. More than 80% of the trace elements in soluble form existed in inorganic and water soluble forms. Conclusion: The morphological characteristics and dissolution characteristics of trace elements provide the theoretical basis for the clinical application of Cortex Moutan.
