从新生小牛的气管灌洗液中用差速离心法提取了肺表面活性物质,其脂质提取物中含磷脂86.1±2.1%(X±SD)、蛋白质1.8±1.1%,以及少量胆固醇、脂肪酸、甘油酯和碳水化合物。磷脂中主要是卵磷脂(83.4±5.6%),其中双饱和卵磷脂占76.0±4.7%。我们用自己研制的改良的Wilhelmy式膜天平测定了提取物的理化特性。最小表面张力是2.4±1.5 mN/m,稳定系数1.78±0.15,当表面张力为10mN/m时,压缩系数0.022±0.004 m/mN,磷脂表面浓度1.112±0.283 nmol/Cm~2。这种提取物能很快吸附到气液界面,并迅速扩展开来。结果表明,我们提取的小牛肺表面活性物质的化学成分和理化特性的各项指标符合替代疗法研究的要求。
Pulmonary surfactant was extracted from neonatal calf tracheal fluid by differential centrifugation. The lipid extract contained 86.1 ± 2.1% (X ± SD) of phospholipids and 1.8 ± 1.1% of protein, and a small amount of cholesterol, Fatty acids, glycerides and carbohydrates. Lecithin was mainly phospholipid (83.4 ± 5.6%), of which double-saturated lecithin accounted for 76.0 ± 4.7%. We measured the physicochemical properties of the extracts using a modified Wilhelmy membrane balance developed by ourselves. The minimum surface tension was 2.4 ± 1.5 mN / m and the stability coefficient was 1.78 ± 0.15. The compressive coefficient was 0.022 ± 0.004 m / mN and the surface concentration of phospholipid was 1.112 ± 0.283 nmol / cm ~ 2 when the surface tension was 10 mN / m. This extract quickly adsorbs to the gas-liquid interface and spreads rapidly. The results showed that the indexes of chemical composition and physicochemical properties of the extracted calf pulmonary surfactant met the requirements of alternative therapy research.
