目的:观察链尿佐菌素(STZ)诱导糖尿病大鼠与非糖尿病大鼠骨组织中TGF-1基因表达变化。方法:将60只Wistar大鼠随机分为正常组、非糖尿病组和链尿佐菌素(STZ)诱导糖尿病组。取腹主静脉血,用Roach P800检测各组大鼠血中的血糖(GLU)、甘油三酯(TG)、低密度载脂蛋白胆固醇(LDL)、总胆固醇(TC)、Ca2+变化;取各组大鼠股骨,用Real-Time RT-PCR(Real-Time Quantitative RT-PCR)方法测定大鼠股骨中TGF-β1 mRNA基因表达变化。结果:(1)与正常组比较,非糖尿病组和II型糖尿病组血中的GLU、甘油三酯(TG)、低密度载脂蛋白胆固醇(LDL)、总胆固醇(TC)含量明显升高(P<0.05),而Ca2+明显下降(P<0.05)。(2)与正常组比较,非糖尿病组和II型糖尿病组离体股骨上1/3骨密度显著降低(P<0.01);(3)与正常组比较,非糖尿病组和II型糖尿病组骨组织TGF-β1 mRNA基因表达上调(P<0.05)。结论:STZ诱导的II型糖尿病模型骨组织中骨密度明显下降,TGF-β1基因表达明显上调,并伴有血脂代谢紊乱,表明II型糖尿病可能通过TGF-β1基因表达上调来促进股骨骨质疏松。
Objective: To observe the changes of TGF-β1 gene expression in streptozotocin (STZ) -induced diabetic rats and non-diabetic rats. Methods: Sixty Wistar rats were randomly divided into normal group, non-diabetic group and streptozotocin (STZ) -induced diabetic group. The abdominal venous blood was collected and the changes of blood glucose (GLU), triglyceride (TG), low density lipoprotein cholesterol (LDL), total cholesterol (TC) and Ca2 + were measured by Roach P800. The rat femur was used to measure the gene expression of TGF-β1 in the femur of rats by Real-Time RT-PCR (Real-Time Quantitative RT-PCR). Results: (1) GLU, triglyceride (TG), low density lipoprotein cholesterol (LDL) and total cholesterol (TC) were significantly increased in non-diabetic group and type II diabetic group compared with those in normal group P <0.05), while Ca2 + decreased significantly (P <0.05). (2) Compared with the normal group, the BMD of the upper third of femur in non-diabetic group and type II diabetic group was significantly lower than that of the normal group (P <0.01); (3) Compared with the normal group, the non-diabetic group and type II diabetic group Tissue TGF-β1 mRNA gene expression was increased (P <0.05). CONCLUSIONS: The bone mineral density (BMD) of STZ-induced type II diabetes mellitus was significantly decreased, and the expression of TGF-β1 was significantly upregulated with dyslipidemia, indicating that type II diabetes may promote osteoporosis of the femur through TGF-β1 gene expression .
