[目的]研究食管鳞状细胞癌组织中miR-34a表达水平及其临床意义。[方法]采用实时反转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)技术检测75例食管鳞状细胞癌组织及配对癌旁组织中miR-34a的表达水平,2-ΔΔCt法计算食管鳞状细胞癌组织中miR-34a相对表达量,进一步结合临床、病理及随访资料进行统计分析。[结果]miR-34a在75例食管鳞癌组织中相对表达量高于癌旁组织;多因素分析显示,吸烟史和临床分期是食管鳞癌组织中miR-34a表达的独立影响因素。miR-34a低表达组的无进展生存期和总生存期均明显劣于高表达组;多因素分析显示miR-34a低表达是无进展生存期和总生存期缩短的独立影响因素。[结论]食管鳞癌组织中miR-34a表达水平可作为预测预后的一项重要指标。
[Objective] To investigate the expression of miR-34a in esophageal squamous cell carcinoma and its clinical significance. [Method] Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of miR-34a in 75 cases of esophageal squamous cell carcinoma and paired paracancerous tissues, 2-ΔΔCt Method to calculate the relative expression of miR-34a in esophageal squamous cell carcinoma tissues, and then further analyze the clinical, pathological and follow-up data. [Results] The relative expression level of miR-34a in 75 cases of esophageal squamous cell carcinoma tissues was higher than that of paracancerous tissues. Multivariate analysis showed that smoking history and clinical stage were the independent influencing factors of miR-34a expression in esophageal squamous cell carcinoma tissues. The progression-free survival and overall survival of miR-34a low expression group were significantly worse than those of high expression group; Multivariate analysis showed that miR-34a low expression was the independent influencing factor of progression-free survival and overall survival shortening. [Conclusion] The expression of miR-34a in esophageal squamous cell carcinoma can be used as an important index to predict the prognosis.
