目的研究RNA干扰抑制结肠癌移植瘤模型VEGF-C表达和抗肿瘤淋巴管生成的有效方法。方法建立结肠癌裸鼠皮下移植瘤模型并随机分为3组(n=8):腺病毒(Ad5F35-VEGF-C-siRNA-EGFP)组、siRNA组和PBS组,对肿瘤进行原位注射,记录各组肿瘤体积变化,免疫组织化学法检测肿瘤内微血管密度(MVD)和淋巴管密度(LVD)。结果VEGF-C干扰能抑制肿瘤生长,且腺病毒组比siRNA组生长更慢。同时免疫组化显示各组MVD值的差异无统计学意义(P>0.05),siRNA组、腺病毒组和PBS组相比,LVD值的差异有统计学意义(P<0.05),LVD值发生不同程度地降低,且腺病毒组降低更显著(P<0.05)。结论病毒介导siRNA干扰VEGF-C的表达是抗肿瘤淋巴管生成的有效方法。
Objective To study the effect of RNA interference on VEGF-C expression and antitumor lymphangiogenesis in colon cancer xenograft model. Methods The subcutaneous xenograft models of colon cancer in nude mice were established and randomly divided into three groups (n = 8): adenovirus (Ad5F35-VEGF-C-siRNA-EGFP), siRNA group and PBS group. Tumor volume changes of each group were recorded. MVD and LVD were detected by immunohistochemistry. Results VEGF-C interference inhibited tumor growth, and adenovirus group grew more slowly than siRNA group. Immunohistochemistry showed that there was no significant difference in MVD between groups (P> 0.05). There was significant difference in LVD between siRNA group, adenovirus group and PBS group (P <0.05), LVD value Decreased to some extent, and adenovirus group decreased more significantly (P <0.05). Conclusions The expression of VEGF-C mediated by siRNA mediated by siRNA is an effective anti-tumor lymphangiogenesis.
