目的探讨染色体分散仪及改良滴片法在染色体制片中的应用。方法使用染色体分散仪控制滴片时环境温湿度,移液枪吸取细胞悬液直接滴加在干净载玻片上;常规消化、染色,观察细胞分散情况;同时按常规方法滴片,计算两种方法获得的染色体分裂相的分散面积比。结果改良滴片法获得的分裂相中染色体呈圆形或椭圆形分散,分布均匀,染色体交叉少,分散面积比高于常规滴片法,差异有统计学意义,变异系数低于常规法。结论通过改良滴片法可制备质量稳定、分布均匀、染色清晰、适于分析的染色体玻片;该法操作简单,效果肯定,建议在各实验室推广。
Objective To explore the application of chromosome disperser and modified droplet method in chromosome preparation. Methods Chromosome scatter was used to control the temperature and humidity in the droplet. The pipette was used to absorb the cell suspension directly on a clean glass slide. The cells were digested and stained routinely to observe the cell dispersion. At the same time, Dispersed area ratio of chromosomal split phase obtained. Results Chromosomes in the split phase obtained by modified droplet method were evenly distributed in circular or oval shape with less chromosome crossover and higher dispersion area ratio than conventional droplet method. The difference was statistically significant and the coefficient of variation was lower than that of conventional method. Conclusion The modified slide method can be used to prepare chromosome slides with stable quality, uniform distribution, clear staining and suitable analysis. The method is easy to operate with definite effect and is recommended to be popularized in various laboratories.
