目的:探讨细胞因子信号传导蛋白STAT3及其抑制因子PIAS3在慢性肾衰竭营养不良大鼠中的表达情况,以及人参养荣汤对其的调节作用。方法:予0.5%腺嘌呤和4%酪蛋白混合饲料喂养制作CRF营养不良大鼠模型,将符合条件的随机分为模型组、人参养荣汤组和开同组,同时设正常组。各组分别予蒸馏水、人参养荣汤(含生药11.43 g/kg.d)、开同(1 g/kg.d)灌胃。治疗4周后检测白蛋白、血红蛋白的变化,Western免疫印迹法检测信号转导蛋白STAT3、PIAS3在下丘脑的表达。结果:人参养荣汤可增加白蛋白、血红蛋白水平,减低血浆瘦素水平,与正常组比较,模型组及人参养荣汤组STAT3表达均增强(P<0.001),人参养荣汤组STAT3表达显著低于模型组(P<0.001);模型组下丘脑PIAS3表达较正常组显著增强(P<0.001);人参养荣汤组PIAS3蛋白表达比模型组有显著增强(P<0.05)。结论:人参养荣汤通过上调CRF营养大鼠下丘脑PIAS3蛋白的表达水平,下调STAT3蛋白表达,从而进一步阻止瘦素受体JAK2/STAT3的信号传导,从而改善CRF营养不良状态。
AIM: To investigate the expression of STAT3 and PIAS3 in malnutrition rats with chronic renal failure and the regulatory effect of Ginseng and Rongrong Decoction on it. Methods: CRF malnutrition rats were fed with mixed diet of 0.5% adenine and 4% casein. The rats were randomly divided into model group, ginseng and nourishing yangtang decoction group and control group with normal group. Each group was given distilled water, ginseng Rong Rong Tang (containing crude drug 11.43 g / kg.d), open the same (1 g / kg.d) gavage. After 4 weeks of treatment, the changes of albumin and hemoglobin were detected. Western blotting was used to detect the expression of STAT3 and PIAS3 in the hypothalamus. Results: Ginseng Yangrong Decoction could increase albumin and hemoglobin level and decrease plasma leptin level. Compared with normal group, STAT3 expression in model group and ginseng yangrong decoction group increased (P <0.001), STAT3 expression in ginseng yangrong decoction group (P <0.001). The expression of PIAS3 in hypothalamus of model group was significantly higher than that of normal group (P <0.001). The expression of PIAS3 protein in ginseng yangrong decoction group was significantly higher than that of model group (P <0.05). Conclusion: Ginseng Yangrong Decoction can improve the malnutrition state of CRF by up-regulating the expression of PIAS3 protein in hypothalamus and down-regulating the expression of STAT3 protein in CRF-fed rats, thereby further preventing the signal transduction of leptin receptor JAK2 / STAT3.
