以氢化大豆磷脂(HSPC)和二棕榈酰磷脂酰胆碱(DPPC)为复合磷脂,制备了马钱子总生物碱复合磷脂脂质体,并与常规的DPPC温敏脂质体比较温敏释放性质。采用浸渍法提取马钱子总生物碱并进行了纯化。采用硫酸铵梯度法和隐形脂质体技术制备了马钱子总生物碱复合磷脂脂质体、温敏脂质体、不具备温敏释放性质的常规脂质体,比较了三者的包封率、粒径、电位、温敏释放行为。3种脂质体的包封率相当,复合磷脂脂质体与温敏脂质体的粒径明显小于常规脂质体,但载体的表面电位没有差别。比较了3种脂质体在37,43℃下释放行为的差别,结果表明,马钱子总生物碱复合磷脂脂质体在37℃的释放明显低于温敏脂质体,而在43℃的释放度高于温敏脂质体,常规脂质体由于相变温度很高,在这2种温度下都几乎不释放。该文的研究结果表明在溶血磷脂用量减少2.5倍的情况下,复合磷脂脂质体具有更好的温敏释放行为。
The total phospholipids of Strychnos nuovars alkaloids were prepared by hydrogenated soybean phospholipid (HSPC) and dipalmitoylphosphatidylcholine (DPPC) as complex phospholipids and compared with conventional DPPC thermosensitive liposomes nature. The results of this study show that the phospholipid liposomes have better temperature-sensitive release behavior when the dosage of lysophospholipid is reduced by 2.5 times.
