目的研究GX-50抗皮肤衰老的作用。方法通过在20月高龄小鼠背部涂抹GX-50,采用HE染色和免疫组织化学法对GX-50抗皮肤衰老的作用进行组织学观察。原代培养人皮肤成纤维细胞,采用H2O2诱导建立皮肤细胞衰老模型,分为空白对照组、DMSO组、GX-50组、H2O2组和GX-50预处理后H2O2处理组(GX-50+H2O2组);采用β-半乳糖苷酶染色实验以及测定细胞分泌物丙二醛(MDA)和细胞内活性氧(ROS)含量检测GX-50在细胞水平的抗衰老能力。应用Draize兔眼刺激性实验评价GX-50的刺激性。结果在组织学水平,GX-50可使高龄小鼠衰老的皮肤年轻化,增加胶原蛋白的表达。在细胞水平,GX-50可使人皮肤成纤维细胞β-半乳糖苷酶合成降低;GX-50组细胞MDA分泌量较空白对照组下降30.45%,GX-50+H2O2组较H2O2组下降61.46%,组间比较差异均有统计学意义(P<0.05);GX-50组细胞胞内ROS荧光值较空白对照组下降8.03%,GX-50+H2O2组较H2O2组下降8.36%,组间比较差异均有统计学意义(P<0.05)。Draize兔眼刺激性评分为0。结论 GX-50可以在一定程度上保护和延缓皮肤衰老,具有作为护肤品有效添加剂的应用前景。
Objective To study the effect of GX-50 on skin aging. Methods GX-50 was applied to the back of 20-month-old mice. The effect of GX-50 on skin aging was observed by HE staining and immunohistochemistry. Primary culture of human skin fibroblast cells was induced by H2O2 to establish skin cell aging model. The cells were divided into blank control group, DMSO group, GX-50 group, H2O2 group and GX-50 pretreatment group Group). The anti-aging ability of GX-50 at the cellular level was determined by β-galactosidase staining assay and the measurement of malondialdehyde (MDA) and intracellular reactive oxygen species (ROS) contents. The irritation of GX-50 was evaluated using a Draize rabbit eye irritation test. Results At the histological level, GX-50 can rejuvenate the senile skin of aged mice and increase the expression of collagen. At the cellular level, GX-50 could reduce the synthesis of β-galactosidase in human dermal fibroblasts; the amount of MDA secreted by GX-50 cells decreased by 30.45% compared with the blank control group, and the GX-50 + H2O2 group decreased by 61.46 (P <0.05). The fluorescence intensity of intracellular ROS decreased by 8.03% in GX-50 group and 8.36% in GX-50 + H2O2 group compared with that of H2O2 group The differences were statistically significant (P <0.05). Draize rabbit eye irritation score is 0. Conclusion GX-50 can protect and delay skin aging to a certain extent, and has the potential application as an effective additive for skin care products.
