目的观察ZEA对离体培养HeLa细胞的生长抑制作用和对超微结构影响,探讨其毒性作用机制。方法用四甲基偶氮唑蓝(MTT)检测不同剂量ZEA暴露24 h后,对HeLa细胞生长抑制作用。在透射电镜下观察ZEA所致细胞超微变化情况。结果 Hela细胞生长抑制率组间比较,差异有统计学意义(F=1 099.093,P<0.05);任何两组间细胞生长抑制率比较,差异有统计学意义(P<0.05);细胞生长抑制率随着染毒剂量的增加而增加。电镜下可见低剂量组细胞胞质内线粒体空泡变性和髓样变;高剂量组坏死细胞数量明显增多,细胞核固缩、畸变、崩解,线粒体嵴絮状变。结论 ZEA对Hela细胞有明显的细胞毒性作用并可导致细胞超微结构损伤。
Objective To observe the growth inhibitory effect and ultrastructure of ZEA on HeLa cells in vitro and to explore the mechanism of its toxicity. Methods MTT assay was used to detect the growth inhibition of HeLa cells exposed to different doses of ZEA for 24 h. The ultrastructural changes induced by ZEA were observed under transmission electron microscope. Results The growth inhibition rates of Hela cells were significantly different between the two groups (F = 1 099.093, P <0.05). There was a significant difference between any two groups in cell growth inhibition rate (P <0.05) The rate of increase with the dose increased. Under the electron microscope, mitochondrial vacuoles and myeloid degeneration were observed in the cytoplasm of low-dose group. The number of necrotic cells in the high-dose group was significantly increased, and the nuclei were pyknotic, distorted, disintegrated and the mitochondrial flocs changed. Conclusion ZEA has obvious cytotoxicity on Hela cells and can lead to the ultrastructure damage of cells.
