目的建立鹿茸和鹿角与其混淆品狍子角、坡鹿角、驯鹿角和驼鹿角的水溶性蛋白的非变性聚丙烯酰胺凝胶电泳(Native-polyacrylamide gel electrophoresis,Native-PAGE)和十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate-polyacrylamide gel electrophoresis,SDS-PAGE)鉴别方法,并对2种电泳方法进行比较。方法采用Native-PAGE和SDS-PAGE两种电泳系统分别对不同产地鹿茸和鹿角与其混淆品水溶性蛋白的组成及其分子量分布范围进行比较研究。结果在NativePAGE图谱中,花鹿茸、马鹿茸、梅花鹿角和马鹿角均具有4条特征谱带,而混淆品狍子角、坡鹿角、驯鹿角和驼鹿角均具有1条特征谱带。正品组与混淆品组间特征谱带大小和位置有明显差异,但鹿茸和鹿角的电泳行为相似。在SDS-PAGE图谱中,花鹿茸、马鹿茸、梅花鹿角和马鹿角的特征谱带数目分别为4条、7条、3条和4条,而混淆品狍子角、坡鹿角、驯鹿角和驼鹿角均具有1条特征谱带,4种鹿茸和鹿角药材及其混淆品的特征谱带的大小和位置存在明显不同。结论 Native-PAGE法不能区别鹿茸与鹿角,但可区分它们与其混淆品药材。而SDS-PAGE法的图谱具有更多鉴别谱带,可明确区分鹿茸与鹿角及其混淆品药材,本研究为鹿茸和鹿角质量综合评价和控制方法提供了科学依据。
Objective To establish Native-polyacrylamide gel electrophoresis (Native-polyacrylamide gel electrophoresis, Native-PAGE) and sodium lauryl sulfate - sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) identification methods, and two kinds of electrophoresis methods were compared. Methods Native-PAGE and SDS-PAGE electrophoresis systems were used to compare the composition and molecular weight distribution of antler, antler and water-soluble protein in different regions of origin. Results In the NativePAGE map, there are four characteristic bands in flower antler, velvet antler, sika antler and red deer antler, while the mixed antler antler, antler antler, reindeer ant and camel ant have one characteristic band. There was a significant difference in the size and location of the characteristic bands between the authentic group and the confused group, but the electrophoresis behavior of the antler and the antler was similar. The number of characteristic bands of antler, antler, sika antler and antler was 4, 7, 3 and 4, respectively, in the SDS-PAGE, while the antlers of antlers, slope antlers, reindeer antlers and moose There are 1 characteristic bands in the angle, and there are obvious differences in the size and location of the characteristic bands of the 4 kinds of pilose antlers and the antler medicinal materials and their confused products. Conclusion Native-PAGE method can not differentiate antler and antler, but they can be distinguished from their confused herbs. However, the SDS-PAGE method has more discriminative bands, which can distinguish the antler from the antlers and their confused herbs. This study provides a scientific basis for the comprehensive evaluation and control of antler and antler quality.
