目的:建立同时测定大鼠血中9个成分(白芍苷、芍药苷、甘草苷、黄芩苷、盐酸小檗碱、汉黄芩苷、甘草酸、黄芩素、汉黄芩素)含量的方法,并将该方法用于清清颗粒的药代动力学研究。方法:采用Promosil C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-0.1%甲酸水溶液梯度洗脱,流速0.3 m L·min~(-1),柱温35℃;采用正离子模式;分为两段时间检测,前7min只检测白芍苷、芍药苷和甘草苷,8~17 min检测其余化合物。结果:各成分的线性关系良好(r>0.998);日间和日内精密度RSD均<15%;准确度的相对回收率90.58%~108.67%。9种成分的达峰时间(tmax)依次为(0.25±0),(0.25±0),(0.25±0),(0.25±0),(1.00±0.10),(6.33±0.82),(1.00±0.11),(0.25±0),(3.12±1.15)h,半衰期(t1/2)依次为(0.21±0.02),(6.72±2.10),(2.70±0.36),(6.90±1.55),(5.16±0.95),(4.34±1.02),(4.74±2.52),(4.79±1.48),(5.78±2.41)h,药峰浓度(Cmax)依次为(3.08±0.94),(10.01±2.17),(18.73±2.84),(36.18±7.57),(50.76±10.61),(793.70±214.08),(54.73±18.01),(207.35±27.58),(50.79±5.95)μg·L~(-1)。结论:该方法简便、准确、重复性好,专属性强,可用于清清颗粒的药代动力学研究。
OBJECTIVE: To establish a method for the simultaneous determination of 9 components in rat blood (white peonyone, paeoniflorin, glycyrrhizin, baicalin, berberine hydrochloride, baicalin, glycyrrhizin, baicalein and wogonin) This method was used to study the pharmacokinetics of clear particles. Methods: The mobile phase was acetonitrile - 0.1% formic acid gradient elution gradient of 0.3 m L · min ~ (-1) with Promosil C18 column (4.6 mm × 250 mm, 5 μm) ; Divided into two time test, only the first 7min detection of white peonyone, paeoniflorin and glycyrrhizin, 8 ~ 17min detection of the remaining compounds. Results: The linearity of each component was good (r> 0.998). The RSD of intra-day and intra-day precision was less than 15%. The relative recoveries of accuracy were 90.58% -108.67%. The peak tmax of the nine components were (0.25 ± 0), (0.25 ± 0), (0.25 ± 0), (0.25 ± 0), (1.00 ± 0.10), (6.33 ± 0.82) (0.21 ± 0.02), (6.72 ± 2.10), (2.70 ± 0.36) and (6.90 ± 1.55), respectively, and the half life (t1 / 2) 5.16 ± 0.95, 4.34 ± 1.02, 4.74 ± 2.52, 4.79 ± 1.48, 5.78 ± 2.41 h, respectively. The peak values of Cmax were (3.08 ± 0.94), (10.01 ± 2.17), (18.73 ± 2.84), (36.18 ± 7.57), (50.76 ± 10.61), (793.70 ± 214.08), (54.73 ± 18.01), (207.35 ± 27.58) and (50.79 ± 5.95) μg · L -1, respectively. Conclusion: The method is simple, accurate, reproducible and specific. It can be used to study the pharmacokinetics of Qingqing granule.
