目的建立高效液相色谱-串联三重四级杆质谱法(HPLC-MS/MS)测定毛樱桃叶中槲皮苷、金丝桃苷、槲皮素、山奈酚、阿福豆苷、山奈酚-3-O-芸香糖苷和芦丁7种主要黄酮类化合物的含量。方法以Agilent ZORBAX SB-C18柱(150 mm×4.6 mm,5.0μm)为分析柱,以乙腈(A)和体积分数为0.1%的甲酸水溶液(B)为流动相体系,梯度洗脱,流速为0.8 m L·min~(-1)。采用电喷雾电离源(electrospray ionization,ESI)负离子化方式检测,以多重反应监测(multiple reaction monitoring,MRM)方式进行扫描定量。结果槲皮苷、金丝桃苷、槲皮素、山奈酚、阿福豆苷、山奈酚-3-O-芸香糖苷和芦丁分别在质量浓度为125~4 000、35~1 120、6.75~216、1.4~44.8、65~2 080、3.75~120、65~2 080μg·L-1内线性关系良好,相关系数(r)均大于0.999,7种成分的加样回收率为96.8%~103.4%(RSD<3%,n=6)。7种黄酮类化合物的含量质量分数分别为5.234%、0.203%、0.053%、0.005%、0.605%、0.011%、1.130%。结论该方法可用于测定樱桃叶中含量差异较大的黄酮类化合物的含量,为樱桃叶的质量控制提供参考依据。
OBJECTIVE To establish a HPLC-MS / MS method for the determination of quercitrin, hyperin, quercetin, kaempferol, aflatoxin, kaempferol-3 -O-rutinoside and rutin seven kinds of flavonoids content. Methods The analytical column was Agilent ZORBAX SB-C18 (150 mm × 4.6 mm, 5.0 μm). The mobile phase consisted of acetonitrile (A) and 0.1% aqueous formic acid (B) 0.8 m L · min ~ (-1). The electrospray ionization (ESI) negative ionization method was used for the detection and the quantitative detection was performed by multiple reaction monitoring (MRM). Results The contents of quercitrin, hyperin, quercetin, kaempferol, aflatoxin, kaempferol-3-O-rutinoside and rutin were detected respectively at the mass concentration of 125-4000, 35-1 120, 216, 1.4 ~ 44.8,65 ~ 2080,3.75 ~ 120,65 ~ 2080μg · L-1, the correlation coefficients (r) were all above 0.999, the recoveries of seven components were 96.8% ~ 103.4 % (RSD <3%, n = 6). The content of seven flavonoids were 5.234%, 0.203%, 0.053%, 0.005%, 0.605%, 0.011% and 1.130% respectively. Conclusion The method can be used to determine the content of flavonoids in cherry leaves with different content, and provide a reference for the quality control of cherry leaves.
