目的研究黄花香茶菜Isodon sculponeatus地下部分的化学成分及其生物活性。方法采用70%丙酮常温提取,硅胶、葡聚糖凝胶和反相色谱法分离纯化,利用现代波谱技术进行结构鉴定;采用MTT法对70%丙酮提取物、醋酸乙酯部位、正丁醇部位等进行细胞毒活性活性测试。结果从黄花香茶菜地下部分醋酸乙酯部位中分离得到14个化合物,分别鉴定为macrophynin E(1)、19-hydroxyferruginol(2)、2α,3α,24-trihydroxyursa-12,20(30)-dien-28-oic acid(3)、山香三萜二烯酸(4)、stigmast-4-en-6β-ol-3-one(5)、豆甾-3β,6α-二醇(6)、松柏醛(7)、邻苯二甲酸二异丁酯(8)、双(2-乙基己基)邻苯二甲酸酯(9)、正十六烷酸(10)、3β-羟基-5α,8α-桥二氧麦角甾-6,22E-二烯(11)、胡萝卜苷(12)、β-谷甾醇(13)、熊果酸(14)。活性研究显示,70%丙酮提取物、醋酸乙酯部位对肝癌细胞SMMC-7721有一定的抑制活性。结论化合物1~10为首次从该种植物中分离得到,化合物2、3、5、6、8、9为首次从该属植物中分离得到。
Objective To study the chemical constituents and biological activities of the underground part of Isodon sculponeatus. Methods 70% acetone was used to extract at room temperature. The constituents were isolated and purified by silica gel, Sephadex LH-20 and reverse phase chromatography. The structures were elucidated by modern spectroscopy. MTT assay was performed on 70% acetone extract, ethyl acetate fraction, n-butanol fraction And other cytotoxic activity test. Results Fourteen compounds were isolated from the ethyl acetate fraction of the roots of Camellia oleifera, and identified as macrophynin E (1), 19-hydroxyferruginol (2), 2α, 3α, 24-trihydroxyursa-12 and 20 (4), stigmast-4-en-6β-ol-3-one (5), stigmasterol-3β, 6α-diol (7), diisobutyl phthalate (8), bis (2-ethylhexyl) phthalate (9), n-hexadecanoic acid , 8α-bridge dioxynil-6,22E-diene (11), daucosterol (12), β-sitosterol (13), ursolic acid (14). Activity studies have shown that 70% acetone extract, ethyl acetate fraction of liver cancer cells SMMC-7721 has some inhibitory activity. Conclusion Compounds 1 ~ 10 were isolated from this plant for the first time. Compounds 2, 3, 5, 6, 8 and 9 were isolated from this genus for the first time.
