目的探讨从血小板源生长因子α(PDGF-α)受体水平阻断对肺血管平滑肌细胞增殖的影响。方法在肺动脉平滑肌细胞(V SM C)培养液中分别给予不同剂量的受体结构域切除的PDGF-α受体腺病毒重组体A d5CM V-PaR tr(ACP),对细胞不同时相的增殖曲线和c-s is原癌基因表达进行测定。结果5 m l/L、10 m l/L剂量ACP对细胞增殖有明显抑制作用。在5 m l/L ACP作用下,加入PDGF-BB不能促进V SM C的增殖。不同剂量的ACP可使c-s is mRNA的表达上调。5 m l/L ACP作用下,加入PDGF-BB可下调c-s is mRNA的表达。结论5 m l/L、10 m l/L ACP能明显抑制肺V SM C的增殖,使c-s is mRNA的表达上调。
Objective To investigate the effect of platelet-derived growth factor-α (PDGF-α) receptor blockade on the proliferation of pulmonary vascular smooth muscle cells. Methods Different concentrations of PDGF-αreceptor adenovirus recombinant A d5CM V-PaR tr (ACP) with different receptor domains were administered to V SM C medium to proliferate cells in different phases Curves and cs is proto-oncogene expression were determined. Results 5 m L / L, 10 m l / L dose of ACP significantly inhibited cell proliferation. Under the action of 5 ml / L ACP, addition of PDGF-BB did not promote the proliferation of VSMC. Different doses of ACP up-regulated c-s is mRNA expression. Under the action of 5 m L / L ACP, the addition of PDGF-BB reduced the expression of c-s is mRNA. CONCLUSION: 5 m L / L and 10 m L / L ACP can significantly inhibit the proliferation of lung V SM C and up-regulate the expression of c-s is mRNA.
