利用自组装单分子层(SAMs)/普鲁士蓝(PB)/壳聚糖(Chit)界面固定葡萄糖氧化酶(GOD),构建酶生物传感器,对传感器在极性非质子有机溶剂如乙腈(AN)、二甲基亚砜(DMSO)和N,N-二甲基甲酰胺(DMF)存在条件下的电化学特性进行了研究,考察了有机溶剂体积分数对传感器响应的影响。结果表明,传感器在50%(体积分数)AN中的电流响应可达到磷酸缓冲溶液(PBS)中的73%。传感器响应速度快、重现性好、抗干扰能力强,展现了高的灵敏度(57.3mA.M.cm-2)。该传感器用于血清标本中葡萄糖的检测,测定结果与市售葡萄糖酶光度测定试剂盒测定结果相吻合。
An enzyme biosensor was constructed by immobilizing glucose oxidase (GOD) with self assembled monolayers (SAMs) / Prussian blue (PB) / chitosan (Chit) , Dimethylsulfoxide (DMSO) and N, N-dimethylformamide (DMF) were studied. The effects of organic solvent volume fraction on the sensor response were investigated. The results show that the current response of the sensor in 50% (volume fraction) AN can reach 73% in phosphate buffered saline (PBS). Sensor response speed, good reproducibility, anti-interference ability, showing a high sensitivity (57.3mA.M.cm-2). The sensor is used for the detection of glucose in serum samples. The results are in good agreement with the results of a commercially available glucose enzymatic photometric kit.
