目的探析白细胞介素7(IL-7)对不明原因复发性流产(URSA)小鼠模型外周血CD4~+/CD8~+T淋巴细胞、Th17/Treg细胞以及妊娠结局的作用。方法选取SPF级实验动物雄性DBA/2和雌性CBA/J小鼠、BALB/c雄鼠。随机将CBA/J小鼠分成流产小鼠组和正常小鼠对照组,又依据有无使用IL-7将流产组分为流产观察组和流产对照组。比较3组小鼠的胚胎吸收率、脾淋巴细胞以及子宫淋巴细胞。结果 3组小鼠的胚胎吸收率组间比较具有显著性差异(P<0.01),且流产观察组小鼠胚胎吸收率最高,正常对照组最低。3组小鼠的CD4~+CD25~+Foxp3~+/CD4~+组间比较均有着显著统计学差异(P<0.01),且流产观察组最低,正常对照组最高;而3组小鼠的CD4~+IL-17A~+/CD4~+组间比较同样显著统计学差异(P<0.01),且流产观察组最高,正常对照组最低。3组小鼠的子宫CD4~+比例、CD4~+/CD8~+组间比较均有显著性差异(P<0.01),且均为正常对照组最高,流产观察组最低。结论对URSA小鼠模型,IL-7可致小鼠外周血CD4~+CD8~+T淋巴细胞、Th17/Treg平衡紊乱,进而影响妊娠结局。
Objective To investigate the effect of interleukin-7 (IL-7) on CD4 ~ + / CD8 ~ + T lymphocytes, Th17 / Treg cells and pregnancy outcome in peripheral blood of URSA mouse model. Methods Male DBA / 2 and female CBA / J mice and BALB / c male mice of SPF class were selected. CBA / J mice were randomly divided into abortion mice and normal mice control group, and then abortion group was divided into miscarriage observation group and abortion control group according to the presence or absence of IL-7. The embryo absorptivity, splenic lymphocytes and uterine lymphocytes in 3 groups of mice were compared. Results The embryo absorption rate of the three groups showed significant difference (P <0.01), and the abortion rate of aborted observation group was the highest, while that of the normal control group was the lowest. The CD4 ~ + CD25 ~ + Foxp3 ~ + / CD4 ~ + groups in three groups had significant statistical difference (P <0.01), and the lowest in abortion group and the highest in normal control group There were also significant differences between CD4 ~ + IL-17A ~ + / CD4 ~ + group (P <0.01), and the abortion group was the highest and the lowest in the normal control group. There were significant differences in CD4 ~ + ratio and CD4 ~ + / CD8 ~ + between the three groups of mice (P <0.01), which were the highest in the normal control group and the lowest in the abortion observation group. Conclusion URSA mouse model, IL-7 can cause CD4 ~ + CD8 ~ T lymphocytes in mice, Th17 / Treg balance disorders, and then affect the pregnancy outcome.
