目的研究黄芪多糖(APS)在支气管哮喘(哮喘)治疗中对气道平滑肌细胞α-平滑肌肌动蛋白(α-SMA)表达的影响及其作用机制。方法由膜荚黄芪提取APS,培养人气道平滑肌细胞系,设APS20、100、500μg/mL3种浓度组及对照组、脂多糖(LPS)组和Toll样受体4(TLR4)拮抗剂组,应用转化生长因子-β(TGF-β)诱导细胞α-SMA的高表达,细胞免疫化学染色、Western blot及PCR技术检测各组的α-SMA、mRNA水平。结果 APS中高浓度组可抑制气道平滑肌细胞α-SMA的高表达(P<0.01),降低α-SMA、mRNA水平(P<0.01),TLR4拮抗剂可部分抑制APS对α-SMA的作用(P<0.01)。结论 APS通过TLR4抑制由TGF-β诱导的气道平滑肌细胞α-SMA的高表达,这可能成为防治哮喘的新途径。
Objective To investigate the effect of astragalus polysaccharides (APS) on the expression of α-smooth muscle actin (α-SMA) in airway smooth muscle cells and its mechanism during the treatment of bronchial asthma (asthma). Methods APS was cultured in Astragalus membranaceus and cultured in airway smooth muscle cell line. APS groups of 20, 100, 500μg / mL and control group, LPS group and Toll-like receptor 4 (TLR4) The expression of α-SMA was induced by transforming growth factor-β (TGF-β), and the level of α-SMA and mRNA was detected by immunocytochemistry, Western blot and PCR. Results High-concentration APS could inhibit the expression of α-SMA (P <0.01), decrease the level of α-SMA and mRNA in airway smooth muscle cells (P <0.01), and TLR4 antagonist could partially inhibit the effect of APS on α-SMA P <0.01). Conclusion APS inhibits the high expression of α-SMA induced by TGF-β in airway smooth muscle cells through TLR4, which may be a new way to prevent and treat asthma.
