目的:探讨线粒体途径在葛根素(PUE)诱导肺动脉平滑肌细胞凋亡中的作用。方法:葛根素高、中、低剂量(1.5×10-3,1.5×10-4,1.5×10-5 mol.L-1)干预体外培养的大鼠肺动脉平滑肌细胞(PASMC),观察线粒体膜电位的变化,Western Blot检测凋亡相关基因Caspase-9,Bax和Bcl-2的蛋白表达。结果:葛根素加药组与正常组相比线粒体膜电位明显降低,葛根素可使Caspase-9的蛋白表达增强,Bcl-2蛋白表达下降而Bax蛋白表达升高,葛根素对PASMC的诱导作用还具有浓度依赖性。结论:葛根素可通过线粒体途径诱导PASMC凋亡。
Objective: To investigate the role of mitochondrial pathway in the induction of pulmonary artery smooth muscle cell apoptosis by puerarin (PUE). Methods: Pulmonary artery smooth muscle cells (PASMCs) were cultured in vitro by high, medium and low doses of puerarin (1.5 × 10-3,1.5 × 10-4,1.5 × 10-5 mol·L-1) Potential changes were detected by Western Blot protein expression of apoptosis-related genes Caspase-9, Bax and Bcl-2. Results: Compared with the normal group, puerarin decreased the mitochondrial membrane potential obviously. Puerarin increased the expression of Caspase-9 protein, decreased the expression of Bcl-2 protein and increased the expression of Bax protein. The induction of PASMC by puerarin It also has concentration dependence. Conclusion: Puerarin can induce apoptosis of PASMC through mitochondrial pathway.
