采用荧光探针技术,通过测定芘荧光发射峰强度的比值I1/I3,研究β-葡聚糖在水溶液中疏水微区的变化情况。试验结果表明,β-葡聚糖溶液在极小质量浓度下基本不形成疏水微区,当质量浓度超过一定值时开始大量形成疏水微区,I1/I3值迅速下降。在相同质量浓度条件下,相对分子质量较小的β-葡聚糖所形成的疏水微区数量也较少。尿素能够破坏疏水微区的形成,氯化钠能够促进疏水微区的形成。
Fluorescence probe technology was used to study the changes of hydrophobic micro-domains of β-glucan in aqueous solution by measuring the ratio of fluorescence emission intensity of pyrene I1 / I3. The results showed that the β-glucan solution did not form hydrophobic micro-domains at very small mass concentration. When the mass concentration exceeded a certain value, a large number of hydrophobic micro-domains began to form, and the value of I1 / I3 decreased rapidly. Under the condition of the same mass concentration, the number of hydrophobic micro-domains formed by β-glucan with relatively small molecular weight is also small. Urea can destroy the formation of hydrophobic microdomains, and sodium chloride can promote the formation of hydrophobic microdomains.
