本文设计一种毛细玻管干冻法以简化菌种保存手续。所用悬浮培养基的成份中含1%谷氨酸钠,5%聚乙烯吡咯烷酮(Polyvinglpyrrolidone),5%蔗糖和1%胰酰糖(tryptose),分装在带螺帽试管(16×125毫米)中,每管2毫升,高压灭菌备用。取内径1.35毫米,外径1.8毫米的毛细玻管,截成25毫米长的一小段,每10根装入一小瓶,用带缝的橡皮塞塞好,高压灭菌备用。待冻干的菌种在固体培养基上培养18—24小时,用无菌棉拭擦下菌苔,放入培养基中,用力振摇使成均匀悬液,将其注入装有10根
In this paper, a capillary tube freeze-drying method to simplify the preservation of species procedures.
