用自制的三系抗重组人α_(2a)型干扰素(rHu-IFN-α_(2a))单克隆抗体,经中和试验,ELISA竞争试验和ELISA夹心试验,对α_(2a)型干扰素抗原表位与抗病毒作用之间的关系,以及干扰素激活靶细胞时与受体作用的方式等进行了研究,并提出了以下设想:(1)α_(2a)型干扰素分子只有一种抗病毒活性区。(2)活性区内至少有2个抗原表位。(3)干扰素抗病毒活性是其整个活性区与靶细胞上受体作用的结果,活性区内任何一个表位都不能单独完成抗病毒的作用。(4)活性区内每个表位对整个活性区的活性都非常重要,缺一不可。
The interferon α 2a interferon (rHu-IFN-α 2a) was detected by neutralization test, ELISA competition test and ELISA sandwich assay using a self-made monoclonal antibody against recombinant human α 2a interferon The relationship between antigenic epitopes and antiviral activity and the way of interferon activation of target cells with receptors are also discussed. The following ideas are proposed: (1) There is only one type of interferon alpha 2a molecule Antiviral activity area. (2) There are at least 2 antigenic epitopes in the active area. (3) The antiviral activity of interferon is the result of the interaction between the entire active region and the receptor on the target cell. No epitope in the active region can completely accomplish the antiviral effect. (4) Each epitope in the active region is very important to the activity of the whole active region, both indispensable.
