应用免疫组织化学链霉菌素生物素技术(Labeled streptavidin biotin method,LSAB)检测30例卵巢上皮性癌患者肿瘤组织中GST-π表达,同时采用三磷酸腺苷(ATP)法测定标本对顺铂(CDDP)、阿霉素(ADM)、足叶乙甙(VP-16)、博莱霉素(BLM)、长春新碱(VCR)、5-氟尿嘧啶(5-FU)和丝裂霉素C(MMC)等7种化疗药物的敏感性。结果:①ATP法测得CDDP对GST-π阴性和阳性肿瘤细胞的平均抑制率分别为(61.1±16.7)%及(43.0±15.1)%(P<0.01);而ADM分别为(48.6±13.6)%及(34.7±15.9)%(P<0.05)。故卵巢上皮性瘸细胞对CDDP和ADM的敏感性与GST-α表达明显相关;而其它5种化疗药物则与GST-π表达无相关(P>0.05)。②ATP法预测化疗敏感或耐药的准确性分别为88.2%或84.6%;GST-π表达预测化疗敏感或耐药的准确性分别为92.8%或75.0%;两法联合预测化疗敏感或耐药的准确性分别为100%和90.9%。
The expression of GST-π was detected by immunohistochemical streptavidin-biotin method (LSAB) in 30 cases of epithelial ovarian cancer. Meanwhile, the expression of CDDP and CDDP were detected by adenosine triphosphate (ATP) Adriamycin (ADM), VP-16, BLM, VCR, 5-FU and MMC, etc. Seven kinds of chemotherapeutic drugs sensitivity. Results: ① The average inhibitory rates of CDDP on GST-π negative and positive tumor cells measured by ATP method were (61.1 ± 16.7)% and (43.0 ± 15.1)% (P <0.01), respectively, while those of ADM were (48.6 ± 13.6) % And (34.7 ± 15.9)% (P <0.05). Therefore, the ovarian epithelial lame cells on CDDP and ADM sensitivity and GST-α expression was significantly related to the other five chemotherapy drugs and GST-π expression was not related (P> 0.05). (2) The accuracy of ATP method in predicting chemosensitivity or drug resistance was 88.2% or 84.6%, respectively. The accuracy of GST-π expression was 92.8% or 75.0% for chemosensitivity or drug resistance respectively. The accuracy was 100% and 90.9% respectively.
